Allicin对DSS诱导的小鼠结肠炎的治疗作用及可能机制

【摘要】 目的:明确Allicin对DSS诱导的小鼠结肠炎的治疗作用并进一步分析可能的机制。方法:将24只8周龄雄性BALB/c小鼠随机分为3组:Control组:阴性对照;DSS组:2.5%DSS诱导结肠炎;Allicin组:2.5%DSS诱导结肠炎+Allicin治疗[10 mg/(kg·d),共7 d]。结果:与DSS组比较,Allicin组疾病活动度评分及肠道炎症评分显著降低(P<0.05)。Allicin治疗组小鼠肠上皮细胞凋亡数目显著低于DSS组。Allicin治疗组小鼠肠黏膜炎症因子(IL-6、IL-1β及TNF-α)水平显著低于DSS组,差异均具有统计学意义(P<0.05)。与DSS组比较,Allicin治疗显著抑制了肠黏膜p-JAK2及p-STAT3的表达(P<0.05)。结论:Allicin对DSS小鼠肠炎具有治疗作用,可能的机制包括抗炎,保护肠上皮细胞及抑制JAK2/STAT3信号通路等。

【Abstract】 Objective: To analyze the therapeutical effect of Allicin for colitis mice induced by DSS and its possible mechanisms. Methods: A total of 24 male mice were randomly divided into Control group,DSS group( 2. 5% DSS,7 days) and Allicin group[DSS treatment and Allicin,10 mg/( kg·d),7 d]. Disease activity index,inflammatory score,TUNEL and Western bolt were performed to analyze the effect of Allicin on colitis induced by DSS. Results: With DSS group,the Allicin group had lower disease activity score and inflammatory score( P < 0. 05). Allicin group had a lower number of intestinal epithelial cell apoptosis than that of DSS group,the difference was statistically significant( P < 0. 05). Western bolt analysis shown that Allicin treatment significantly depressed the expression of p-JAK2 and p-STAT3 in intestinal mucosa when compared with these of DSS group( P < 0. 05). Conclusion: Conclusion Allicin has significantly therapeutic effect on mice colitis induced by DSS,the possible mechanisms including anti-inflammatory effects,protected of the intestinal epithelial cells and inhibition of the JAK2 / STAT3 signaling pathways.
