黄芪多糖对甲流HA2蛋白真核表达载体免疫效果影响的研究
Research of astragalus polysaccharides increasing immune effect of influenza A virus HA2 eukaryotic expression vector
【摘要】 目的:检验不同剂量黄芪多糖对甲流HA2蛋白真核表达质粒免疫大鼠效果的影响。方法:首先,将编码HA2的质粒p HA2/EGFP转染CHO细胞,通过PT-PCR和荧光检验其在真核细胞中的表达效果。然后将60只大鼠随机分为6组,分别注射p EGFP-N1、生理盐水和p HA2/EGFP加不同浓度的黄芪多糖,注射前收集大鼠血清作为对照,据最后一次注射后第36天处死各组大鼠收集各组大鼠的血清,测量血清中细胞因子IFN-γ、IL-4和血清中Ig G对不同流感毒株血凝素的反应情况。结果:p HA2/EGFP+中等剂量APS组的IFN-γ、IL-4和血清中Ig G对血凝素的反应情况均与低剂量组和阴性对照组有显著性差异(P<0.05)。p HA2/EGFP+中等剂量APS组的细胞因子水平和对血凝素反应与高剂量组无显著性差别(P>0.05)。结论:中等剂量的黄芪多糖能增强甲流HA2蛋白真核表达质粒的免疫效果。
【Abstract】 Objective: To study the astragalus polysaccharides( APS) effect on immune induced by influenza A virus HA2 eukaryotic expression vector. Methods: The HA2 encoded by the DNA vaccine vector was efficiently expressed in CHO cells,as determined by reverse transcription polymerase chain reaction( RT-PCR) and fluorescence analysis. 60 rats were divided into six groups randomly,which were immunized with normal saline,p EGFP-N1,p HA2 / EGFP +different dose of APS by intramuscular injection. The control sera were collected before injection. After injected the 36 th day,sera were collected to analyzing IFN-γ,IL-4 and Ig G level. Results: IFN-γ,IL-4 and Ig G level of p HA2 / EGFP +m APS group was different from that of p EGFP-N1 group or p HA2 / EGFP +l APS group( P<0. 05). Conclusion: Middle dose of APS could increase immune induced by influenza A virus HA2 eukaryotic expression vector.
