p-P38MAPK与TGF-β1在腰椎间盘退变组织中的表达研究
THE EXPRESSION OF P-P38MAPK AND TGF-Β1 IN THE INTERVERTEBRED DISC DEGENERATED
【摘要】 目的检测正常及退变腰椎间盘组织中p-P38MAPK、TGF-β1的表达及含量,分析二者的相关性。方法选取唐山市第二医院脊柱外科手术切除的86例腰椎间盘突出症患者的椎间盘退变组织(病变组);同时选取12例因腰椎外伤手术切除的正常椎间盘组织(对照组)。采用HE染色、免疫组织化学方法和ELISA方法检测标本中p-P38MAPK、TGF-β1的表达及含量。结果免疫组化实验检测结果病变组椎间盘组织p-P38MAPK、TGF-β1的表达量明显高于对照组,P<0.05。ELISA方法检测结果病变组椎间盘组织p-P38MAPK、TGF-β1的含量明显高于对照组,P<0.05。结论 p-P38MAPK与炎症、应激反应的调控密切相关,其参与了TGF-β1等炎性应激因子的作用过程,认为是椎间盘退变的重要原因之一。
【Abstract】 Objective To investigate the expression of p-P38 MAPK and TGF-β1 and their significance in the disc degeneration tissues.Methods The degenerative disc tissue surgical resection from86 cases with lumbar disc herniation from december 2008 to december 2010 were selected(lesion group);at the same time,normal disc tissue surgical resection from 12 cases trauma were selected(control group).pP38 MAPK and TGF-β1 in content were detected by immunohistochemistry and ELISA,the relations andsignificance of p-P38 MAPK,TGF-β1 with disc degeneration were evaluated.Results Immunohistochemical results showed the expression of p-P38 MAPK and TGF-β1 in lesions group was significantly higher than that in control group,the positive is that brown granules can be seen in cytoplasm,OD value in lesions group were significantly higher,P<0.05.ELISA results showed the content of p-P38 MAPK and TGF-β1in lesions group was significantly higher,P<0.05.Conclusions In the disc degeneration,p-P38 MAPK is an important mitogen activated protein kinase,thus played its key role in disc degeneration.It has broad and complex biological effects.We conside p-P38 MAPK involving inflammation,stress response is closely related to their involvement in the regulation of TGF-β1 and other inflammatory stress factor.The inflammatory factor is one of the important cause at disc degeneration.
