大肠埃希菌ELISA检测条件的优化
Optimization of ELISA Conditions for Escherichia Coli Detection
【摘要】 目的对ELISA检测大肠埃希菌方法条件进行优化研究.方法以大肠埃希菌为对象,通过Box-Behnken设计星点响应面实验进行优化实验.结果优化后ELISA检测条件:抗原包被浓度为6.25μg/m L,抗原包被最佳时间为7 h,抗体稀释度最佳为1∶625 0,大肠埃希菌ELISA检测OD值为2.12.结论经实验得证,通过星点响应面实验方法进行大肠埃希菌ELISA检测条件优化,结果合理有效,优化效果良好.
【Abstract】 Objective To optimize the test conditions of ELISA method for detecting E.coli.Method The ELISA method for detecting E.coli was optimized through Box-Behnken central composite design and response surface methodology.Results The optimized ELISA test conditions were as follows: antigen coating concentration of 6.25 μg/m L antigen coating time of 7 hours,antibody dilution of 1 ∶ 625 0.Under this condition,the detected OD value of E.coli reached 2.12.Conclusion The optimization of ELISA conditions for E.coli detection using the Box-Behnken statistical experimental design is not only scientific and rational,but also quick and efficient.
