盐藻油微波辅助提取工艺优化及脂肪酸组成分析
Optimization of Microwave-Assisted Extraction Technology of Oil from Dunaliella Salina and Its Fatty Acid Component
【摘要】 对微波辅助乙酸乙酯萃取盐藻油工艺进行了优化研究。采用二次通用旋转组合实验设计分析了液固比、浸提温度和浸提时间对盐藻油得率的影响,并建立了相应的预测模型。方差分析结果表明,液固比(P<0.005)、浸提温度(P<0.005)和浸提时间(P<0.005)都对盐藻油脂得率有显著影响,各因素间不存在交互作用,3个因素的影响顺序为:浸提时间>浸提温度>液固比。最佳工艺条件为:液固比13 mL/g,浸提温度78℃和浸提时间12.5 min,在该条件下,盐藻油得率为(16.01±1.15)%。微波辅助提取的盐藻油中主要含有棕榈酸、棕榈油酸、油酸、亚油酸和亚麻酸等脂肪酸,同样以乙酸乙酯为提取溶剂,与常规回流提取和超声波提取相比,微波辅助提取显著增加了盐藻油中亚麻酸的含量(P<0.005),对其它脂肪酸含量没有显著影响(P>0.005)。
【Abstract】 Technology for microwave- assisted oil extraction produced ethyl acetate from Dunaliella salina was investigated.Quadratic rotation combination desiglt was applied to optimize some operation parameters on the effects of microwave- assisted extraction of oil from Dunaliella salina on oil yield.Ratio of liquid to solid,extracting time and temperature affected oil yield significantly(P < 0.005) by t- test and P- value;there was no interaction effect between each parameters(P <0.05);sequence of the effect of three factors on oil yield was as follows;that extraction time > temperature >ratio of liquid to solid.The optimized condition were as follows:ratio of liquid to solid 13mL/g,extracting temperature 78 ℃,extracting time 12.5 min,oil yield of Dunaliella salina(16.01 ±1.15)%in canse that it was extracted with the optimized conditions.The main component of Dunaliella salina oil were:palmitic acid,palmitoleic acid,oleic acid,linoleic acid and linolenic acid etc.,on condition that ethyl acetate was applied as solvent,compared with method of conventional reflux extraction methods and ultrasound assisted extraction,linolenic acid content of Dunaliella salina oil were improved significantly assisted by microwave- assisted extraction method(P < 0.005);the selected extraction methods had no effect on the other fatty acid content significantly(P > 0.05).
