卡铂诱导卵巢癌耐药特性的体外研究

【摘要】 目的通过建立人卵巢癌对卡铂耐药细胞株,研究其耐药特性及耐药后吲哚胺2,3双加氧酶(IDO)的表达,初步探讨耐药与IDO的关系,进一步明确耐药卵巢癌转移或复发的机制。方法模仿临床卵巢癌化疗模型,采用中等剂量、间歇作用法,建立卵巢癌SKOV3细胞对卡铂耐药的细胞系SKOV3/CBP,采用MTT法检测SKOV3和SKOV3/CBP两组细胞的增殖能力,Matrigel小室检测两组细胞的侵袭能力,ELISA法SKOV3/CBP检测两组细胞IDO的表达。结果随着培养时间的延长,两组细胞的增殖力逐渐增加,但SKOV3/CBP细胞的增殖力较SKOV3细胞明显降低,且48 h后两组的增殖力有明显差异(P<0.05);SKOV3/CBP细胞较SKOV3细胞侵袭能力明显增加且有统计学差异(P<0.05),同时IDO的表达在SKOV3/CBP细胞组较SKOV3细胞中增多且有统计学差异(P<0.05)。结论对卡铂耐药的卵巢癌转移主要通过癌细胞的侵袭而非增殖发生;IDO可能作为卵巢癌耐药研究的另一突破点。

【Abstract】 Objective The purpose of this study is to establish carboplatin-resistance cell line, investigate the drug resistance mechanism and expression of indoleamine 2, 3-dioxygenase of drug resistance ovarian cell lines SKOV3. Discussion was made about its relationship of drug resistance and IDOr. Methods The carboplatin-resistance cell line, SKOV3/CBP, was induced by intermittent effect of medium dose carboplatin; The proliferation of SKOV3 and SKOV3/CBP was tested by MTT; The invasion ability of SKOV3 and SKOV3/CBP was tested by Matrigel Plug; the level of indoleamine 2, 3-dioxygenase was measured by immunoassay KIT. Results The proliferation of two groups cells increased gradually with the extension of culture, but the proliferation of SKOV3/CBP cell was less decreased than SKOV3 cell, and there was significantly different proliferation between two cells after culture for 48 hours(P<0.05). The invasion ability of SKOV3/CBP cell significantly increased compared with SKOV3 cells(P<0.05). Moreover, the expression of IDO in SKOV3/CBP cell group was more increased significantly than SKOV3 cells(P<0.05). Conclusion The metastasis of carboplatin resistant ovarian was occurred by cell invasion, rather than cell proliferation; IDO can be used as another breakthrough point for the study of drug resistance of ovarian cancer.
