SMTNL2基因在肾癌组织及细胞系中的表达特征研究
Expression of SMTNL2 in renal cell carcinoma
【摘要】 目的探讨SMTNL2基因在肾癌组织及肾癌细胞系中的表达特征和临床意义。方法采用RT-PCR方法检测SMTNL2 m RNA在肾癌患者肿瘤组织及配对癌旁正常组织的表达及在5种肾癌细胞系(ACHN、Caki、769-P、786-O、293)中的表达,实时荧光定量PCR方法检测SMTNL2 m RNA在36例肾癌组织及配对癌旁正常组织中的表达水平,Western blot和免疫组化方法检测SMTNL2蛋白在36例肾癌组织及配对癌旁正常组织中的表达水平,分析SMTNL2基因在肾癌组织中的表达特征和临床病理参数的相关性。结果 SMTNL2 m RNA在肾癌组织中的表达水平显著低于癌旁正常肾组织(0.089±0.135 vs.0.325±0.165,P<0.05),SMTNL2 m RNA在肾癌细胞系(ACHN、Caki、769-P、786-O、293)中同样表现为低表达或无表达。SMTNL2在肾癌组织中的阳性蛋白表达率明显低于癌旁正常组织(13.9%vs.77.8%,P=0.021)。SMTNL2在肾癌组织中的SMTNL2阳性蛋白表达率与患者的性别(P=0.659)、年龄(P=0.462)、病理分级(P=0.391)和分期(P=0.696)无相关性。结论 SMTNL2基因在肾癌组织及肾癌细胞系中低表达,可能与肾癌的发生和进展有关。
【Abstract】 Objective To investigate the expression of SMTNL2 in renal cell carcinoma and cell lines. Methods The expression of SMTNL2 was detected by RT-PCR, quantitative Real-time PCR, Western blot in renal cell carcinoma tissues, paired adjacent normal tissues, five kinds of renal cancer cell lines(ACHN, Caki, 769-P, 786-O, 293). Western blot and immunohistochemistry staining were used to detect the expression of SMTNL2 protein in 36 cases renal caner tissues, then analyzed the relationship between SMTNL2 expression and clinical characteristics in 36 cases of renal cancer. Results The expression of SMTNL2 in renal cancer tissues was significantly lower than adjacent normal tissues, and it showed no expression or low expression in the 5 kinds of renal cancer cell lines, ACHN, Caki, 769-P, 786-O, 293. The expression of SMTNL2 m RNA in renal cancer tissues was significantly lower than that in adjacent normal tissues(0.089±0.135 vs. 0.325±0.165, P<0.05); The positive expression rate of SMTNL2 in renal cancer tissues was significantly lower than that in normal renal tissue 77.8%(P=0.021). There were no relation among sex(P=0.659), age(P=0.462), grade(P=0.391) and TNM stage(P=0.696) about the positive expression rate of SMTNL2 in renal cancer. Conclusion SMTNL2 genes is low expression in renal cancer tissues and 5 kinds of renal cancer cell lines, SMTNL2 gene may play an important role in the tumorigenesis of renal cell carcinoma.
