染铅大鼠脑组织JWA mRNA表达变化与氧化应激损伤的关系

【摘要】 目的探讨铅暴露对大鼠大脑皮质、小脑、海马组织JWA mRNA表达的影响及其与脑组织氧化应激损伤的关系。方法 40只刚断乳雄性SD大鼠,按体重随机区组法分为5组(对照组和4个乙酸铅剂量组),各组自由饮用不同浓度的乙酸铅溶液(0、100、200、400、800 mg/L)。连续染毒60 d后取大鼠脑组织,RT-PCR技术检测脑组织JWA mRNA的表达量,并测定脑组织铅含量、过氧化氢(H2O2)水平、丙二醛(MDA)及8-羟基脱氧鸟苷(8-OHd G)含量。结果与对照组相比,各染铅组大鼠大脑皮质、小脑和海马组织JWA mRNA表达水平明显降低,而脑组织铅含量及H2O2、MDA及8-OHd G水平均增加(P<0.05);脑组织JWA mRNA的表达量与其铅含量、H2O2、MDA、8-OHd G水平呈负相关(P<0.05)。结论饮水铅暴露可导致大鼠脑组织JWA mRNA的表达变化,并与脑组织氧化应激损伤密切相关。

【Abstract】 Objective To explore the effects of lead exposure on the expression of JWA mRNA in cerebral cortex,cerebellum and hippocampus of rats and the relationship with oxidative stress and damage in brain tissues.Methods 40 weaned male SD rats were divided randomly into five groups according to the weight block design,and drank unlimitedly distilled water containing 0,100,200,400,800 mg/L lead acetate for 60 days respectively. Then the expression of JWA mRNA in the brain was detected by RT-PCR,and the levels of lead,Hydrogen peroxide( H2O2),malondialdehyde(MDA) and 8-hydroxyl deoxyguanosine(8-OHd G) in brain tissue were determined by the related kits respectively.Results Compared with those of control group,the expression of JWA mRNA in cerebral cortex,cerebellum and hippocampus of lead exposure groups was decreased significantly,whereas contents of lead,H2O2,MDA and 8-OHd G in brain tissue were increased( P<0.05).Correlation analysis results revealed that the expression of JWA gene mRNA was negatively correlated with contents of lead,H2O2,MDA and 8-OHd G in brain tissue(P<0.05).Conclusions Exposure to lead through drinking water could affect the expression of JWA mRNA and its expression intimately correlates with oxidative and stress damage of brain tissues.
