鸟类AhR报告基因法对垃圾焚烧炉飞灰中的类二噁英污染物的评估

【摘要】 采用鸟类AhR报告基因法(luciferase reporter gene assay,LRG)评估了典型飞灰样品有机提取物的类二噁英生物活性。结果显示:垃圾焚烧飞灰样品的有机提取物能诱导显著的类二噁英活性,且在总类二噁英的毒性当量值(toxic equivalents,TEQs)上表现出显著的物种间差异,表明了其对环境样品毒性进行多物种评估的必要性。与化学仪器法相比,化学仪器测得飞灰样品含有的17种二噁英物质的Chem-TEQs分别占鸡、环颈雉、日本鹌鹑AhR-LRG法测得的总类二噁英活性Bio-TEQs的0.21%~0.70%、0.10%~0.43%、0.02%~0.11%。该结果表明:对17种二噁英物质的化学仪器暴露分析不能全面反映环境介质中毒害化学物质的类二噁英毒性情况;而鸟类AhR-LRG法不仅能反映样品的总类二噁英活性,还可通过多种物种实现对样品较全面的生态风险评估。

【Abstract】 The activity of dioxins of the typical fly ash samples was evaluated by the avian AhR luciferase reporter gene(AhRLRG)assay.The results indicatethat the dioxin activity of fly ash organic extracts can be detected by the avian AhR-LRG assay,and toxic equivalents(TEQ)of fly ash samples are very different among birds,which show the necessity of assessing the toxicity of environmental samples among different species.Compared to chemical method,the Chem-TEQs values of the fly ash samples are only 0.21-0.70%,0.10-0.43%,0.02-0.11% of Bio-TEQs from AhR-LRG method in the chicken,the pheasant and the quail,respectively.The results show that Chem-TEQs by chemical method based on 17 chemicals can not fully reflect the total dioxin activity of environmental samples.However,the avian AhR-LRG assay can evaluate the total dioxin activity of environmental samples among different species.
