腺相关病毒介导KLF7对小鼠坐骨神经缺损后感觉功能的作用

【摘要】 [目的]探讨腺相关病毒2(AAV2)介导核转录因子KLF7对脱细胞异体神经支架(ANA)修复小鼠坐骨神经缺损后感觉轴突再生和感觉功能恢复的作用。[方法]成年C57BL/6小鼠随机分为正常组、ANA+AAV2对照组和ANA+AAV2-KLF组,术后4周RT-PCR和免疫荧光染色检测神经支架内KLF7 mRNA和蛋白表达,免疫荧光染色检测神经支架内NF和S100蛋白表达,霍乱毒素亚单位B结合荧光素TRITC(CTB-TRITC)逆行示踪L2~5脊髓背根神经节神经元(DRG)标记感觉轴突再生,Hargreaves法和电生理检测感觉功能的恢复。[结果]与对照组比较,ANA+AAV2-KLF组神经支架内KLF7mRNA和蛋白表达显著增高,NF和S100蛋白表达增加,CTB阳性标记DRG感觉神经元数量增加,Hargreaves热缩足反射潜伏期降低,电生理神经传导速度增加,潜伏时缩短,动作电位波幅增大(P<0.05)。[结论]KLF7可促进小鼠坐骨神经缺损后感觉轴突再生和髓鞘形成,促进感觉功能的恢复。

【Abstract】 [Objective]To study the effect of AAV2-KLF7 on sensory axonal regeneration and sensory functional recovery after acellular nerve allograft( ANA) repair of the sciatic nerve gap in rats. [Method]Adult C57 BL /6 mice were randomly divided into Sham group( n = 2),ANA + AAV2 control group( n = 12) and ANA + AAV2-KLF group( n = 12). Four weeks later,KLF7 was detected with RT-PCR and immunohistochemistry in the regenerated nerves. Axonal regeneration and myelination were examined with NF and S100 immunohistochemistry,sensory axonal regeneration were observed with TRITC labeled cholera toxin B( CTB-TRITC) retrograde neural tracing. Meanwhile,sensory function recovery was examined with Hargreaves test and electrophysiological analysis. [Result]Compared with the results from control group,the mRNA and protein expression of KLF7 increased in the regenerated nerves in ANA + AAV2-KLF group. KLF7 overexpression provided several beneficial effects,such as an increase in the amplitude,nerve conduction velocity and enhanced expression of NF,S100 and CTB-labeled neurons in DRG. Furthermore,the latency period shortened in Hargreaves test( P < 0. 05). [Conclusion]Our findings suggest that KLF7 could promote peripheral nerve sensoryaxonal regeneration,myelination formation as well as sensory function recovery.
