AEG-1、NF-κB在胃癌组织中的表达及临床意义

【摘要】 目的探讨AEG-1、NF-κB在胃癌组织中的表达情况及其与胃癌临床病理特征的关系。方法采用免疫组织化学法检测84例胃癌患者AEG-1、NF-κB表达情况,观察两个指标之间的相互关系及其与患者临床病理特征及预后的关系。使用SPSS16.0统计软件进行统计分析,计量数据使用均数±标准差(x珋±s)表示,采用方差分析进行多组间比较,两组间比较采用独立样本t检验;Spearman等级相关分析AEG-1、NF-κB表达的相关性,用COX回归分析模型进行多因素分析,P<0.05为差异具有统计学意义。结果胃癌组织中AEG-1的阳性表达率为75.00%,其阳性表达的PU值明显高于癌旁胃组织(t=12.206,P<0.01);其阳性表达与分化程度、TNM分期、淋巴结转移有关(P<0.05)。NF-κB在胃癌组织中的阳性表达率为80.95%,其阳性表达的PU值明显高于癌旁胃组织(t=11.997,P<0.01);其阳性表达与分化程度、浸润深度、TNM分期、淋巴结转移有关(P<0.05)。胃癌组织中AEG-1、NF-κB阳性表达呈正相关。COX模型多因素分析显示,AEG-1表达情况、淋巴结是否转移及临床TNM分期是影响胃癌术后生存率的指标中最为重要的3个指标(P<0.05)。结论胃癌组织中可能存在AEG-1、NF-κB信号通路。检测胃癌组织中AEG-1、NF-κB的表达,有利于判断胃癌的发生、发展、浸润、转移并可评价胃癌的预后。

【Abstract】 Objective To investigate the expression of AEG-1 and NF-kappa B in gastric cancer tissue and to discuss its relationship with clinical-pathological features of gastric cancer. Methods The expression of AEG-1 and NF-kappa B in 84 patients with gastric cancer was detected using the immunohistochemical staining method. The association of AEG-1 and NF-kappa B in gastric carcinoma and their clinical pathological features and prognosis of the patients were analyzed. Statistical analysis was made using SPSS 16. 0 software. Measurement data were expressed as mean ± SD and were examined using variance analysis and independent sample t test between the 2 groups. The association of AEG-1 / MTDH and NF-kappa B was analyzed using Spearman’s correlation analysis. Multivariate analysis was performed with COX regression analysis model. P < 0. 05 was considered statistically significant. Results The positive expression rate of AEG-1 in cancer tissues was 75. 00%,and the PU value of the positive expression of AEG-1 in gastric cancer was significantly higher than that in paracancer tissue( t = 12. 206,P < 0. 01). The positive expression of AEG-1 in gastric cancer was closely correlated with tumor differentiation degree,TNM staging and lymph node metastasis( P < 0. 05). The positive expression rate of NF-kappa B in gastric cancer tissues was 80. 95%,and the PU value of the positive expression of NF-kappa B in gastric cancer was higher than that in paracancer tissue( t = 11. 997,P < 0. 01). The positive expression of NF-kappa B in gastric cancer was closely correlated with tumor differentiation degree,invasion depth,TNM staging and lymph node metastasis( P < 0. 05). Spearman’s correlation analysis showed that there was a line relationship between AEG-1 and NF-kappa B in gastric cancer. COX regression analysis of the multi-factor model showed that the expression of AEG-1,lymph node metastasis,and TNM staging were the three independent risk factors for postoperative survival of patients with gastric carcinoma( P < 0. 05). Conclusion AEG-1 and NF-kappa B signaling pathway might be involved in gastric carcinoma. The detection of the expression of AEG-1 and NF-kappa B in gastric cancer tissues could help to predict the occurrence,development,invasion and metastasis of gastric carcinoma in addition to the the evaluation of prognosis.
