牛病毒性腹泻病毒LJ36/14的分离与鉴定

【摘要】 为确定牛呼吸道传染病的病原,本研究对2014年从黑龙江省发生牛呼吸道传染病的某肉牛场采集的40份鼻拭子进行了病毒分离,结果分离到一株牛病毒性腹泻病毒(BVDV),命名为LJ36/14株。病毒鉴定结果表明,分离株在单层MDBK细胞中不产生细胞病变;利用BVDV p80蛋白的单克隆抗体(MAb)进行的间接免疫荧光试验表明接种LJ36/14株的细胞结果显示阳性。分离株的TCID50为105.9/0.1 mL。对该分离株的5’端非编码区(5’UTR)和Npro基因进化树分析表明该分离株为国内首次分离并鉴定的基因亚型,即BVDV1v亚型。本研究为了解BVDV地理分布和流行情况及BVDV疫苗的制发奠定了基础。

【Abstract】 To investigate the pathogens related with bovine respiratory disease, forty bovine nasal swab samples were collected from affected cattle with respiratory signs from Heilongjiang province in 2014 and subjected to virus isolation in MDBK cells. A bovine viral diarrhea virus(BVDV) was isolated and identified with the monoclonal antibody against BVDV by indirect immunofluorescence assay, named LJ36/14. The virus was a noncytopathogenic and unable to produce cytopathic effects in MDBK cells. The phylogenetic analysis based on the 5’UTR and Npro gene sequences of LJ36/14 and other reference BVDV strains showed that LJ36/14 was classified into group I and subgenotype v, and so far, the genotype 1v of BVDV was first reported in China, which provided additional information for different subgenotype of BVDVs prevalence in China.
