2009年~2012年内蒙古地区狂犬病毒分离株遗传变异分析与免疫保护效果研究

【摘要】 为了解内蒙古自治区狂犬病毒(RABV)流行株的遗传进化情况,本研究于2009年2012年,在内蒙古地区分离得到16株RABV,测序后获得16条N基因和G基因序列。序列分析表明N基因核苷酸相似性为86.5%100%,G基因核苷酸相似性为83.9%100%。N基因和G基因核苷酸遗传进化分析显示本次分离到的病毒株均属于基因I型,其中12株G蛋白V332I突变属于亚洲Asian谱系,其他4株G蛋白T36A突变属于Cosmopolitan谱系,为内蒙古主要流行的两个谱系。CTN-1(Asian谱系)和ERA(Cosmopolitan谱系)疫苗免疫BALB/c小鼠对分离株NM4(Asian谱系)和NM8(Cosmopolitan谱系)的攻击均能够提供有效保护。本研究初步证实了目前疫苗能够有效地保护人和动物对RABV的感染。

【Abstract】 To investigate the prevalence and evolutionary of rabies virus(RABV) in Inner Mongolia, a total of 16 RABVs were isolated from different areas of Inner Mongolia from 2009 to 2012, and the N genes and G genes of the isolates were sequenced. Sequence analysis showed that the nucleotide similarity of N and G genes of the isolates were 86.5% to 100% and83.9% to 100%, respectively. Phylogenic analysis based on the sequences of N and G gene showed that the isolates belong to the genotype I. Of which 12 isolates with V332 I mutation in G protein were classified into the Asian lineage and other 4 isolates with T36 A mutation in G protein gene belonged to the Cosmopolitan lineage. However, CTN-1(Asian lineage) and ERA(Cosmopolitan lineage) vaccine were able to provided completely protection for the BALB/c mice when challenged with either the isolate NM4(Asian lineage) or NM8(Cosmopolitan lineage). In conclusion, the current vaccines still have the abilities to effectively protect the both human and animals against RABV infection.
