南瓜子叶节再生体系的建立
The regeneration system from cotyledonary node of Cucurbita maxima
【摘要】 为建立南瓜子叶节再生体系,试验以‘上桔’‘金栗’‘银栗’‘贝栗灰’‘永安2号’和‘永安5号’共6个南瓜品种的子叶节为材料,以MS培养基为基本培养基,通过设置9种芽分化培养基,研究6-BA和IBA组合、KT对南瓜的子叶节诱导芽的影响。结果表明,1.0 mg·L-16-BA和0.02 mg·L-1IBA质量浓度组合是6-BA与IBA浓度组合中不定芽诱导率最高的,为66.67%。在培养基中添加2 mg·L-1和3 mg·L-1的KT可以显著提高不定芽的诱导率;基因型不同南瓜的子叶节出芽率不同,其中南瓜品种‘银栗’最易诱导出芽,诱导率为93.75%,而‘上桔’的诱导率最低,诱导率为47.92%。试验得出南瓜子叶节诱导芽的最佳诱导培养基为MS+3 mg·L-1KT+1.0 mg·L-16-BA+0.02 mg·L-1IBA,平均诱导率为83.33%。
【Abstract】 In order to establish the regeneration system of pumpkin cotyledonary node and study the effect of 6-BA,IBA,KT on bud regeneration from pumpkin cotyledonary node,‘Shangju’‘Jinli’‘Yinli’‘Beilihui’‘Yongan No. 2’ and ‘Yongan No. 5’ were used as materials with MS as basic medium and setting up 9 kinds of bud differentiation media. The results showed that the medium with 1.0 mg·L-16-BA and 0.02 mg·L-1IBA combination has the highest rate of adventitious bud induction of the combination and the induction rate was 6.67%. By adding 2 mg·L-1or 3 mg·L-1KT could significantly improve the induction rate of adventitious buds. The adventitious bud induction varied in pumpkin genotypes.‘Yinli’ was the cultivar most easily to induce bud and the induction rate was 93.75%. The induction rate of ‘Shangju’ was the lowest with 47.92%. It was concluded that the best induction medium for the bud induction of pumpkin was MS+3 mg·L-1KT+1.0mg·L-16-BA+0.02 mg·L-1IBA. The average induction rate was 83.33%.
