2013-2014年大连市食源性疾病主动监测中分离的副溶血性弧菌病原特征

【摘要】 目的了解2013-2014年本地区副溶血性弧菌的病原学特征,为临床用药提供依据;同时与各地优势脉冲场凝胶电泳(PFGE)型别进行比较,揭示其流行与发展趋势的规律。方法依据GB/T 4789.7-2008进行副溶血性弧菌生化鉴定,同时用荧光定量PCR方法进行复核;采用微量稀释法进行药敏实验;菌株基因组DNA经限制性内切酶Sfi I酶切,用PFGE方法获得电泳图谱,利用Bio Numerics软件对图谱进行同源性分析。结果 88株副溶血性弧菌荧光定量PCR均检出副溶血性弧菌核酸;所有菌株对8种抗生素均敏感;88株菌的DNA指纹图谱可大致分为38种PFGE图谱,经Bio Numerics软件分析分为A~H 8个群,其中F群为优势群。结论荧光定量PCR方法与常规培养法结合是缩短副溶血性弧菌检出时间、提高检出率的有效方法;从大连市患者粪便中检出的副溶血性弧菌对临床常用的8种抗生素尚无耐药情况且亲缘关系较近。

【Abstract】 Objective To analyze the pathogenic characteristics of Vibrio parahaemolyticus from foodborne diseases active surveillance in Dalian city from 2013 to 2014. To provide basis for clinical medication. To compare the PFGE types with other cities and reveal its regularity of epidemiology and development trend. Methods API biochemical identification was used according to GB / T 4789. 7- 2008 and real- time fluorescence quantitative PCR was used to check the results. Drug sensitive test was carried out by microdilution method. Molecular typing of strains was analyzed by pulse- field gel electrophoresis( PFGE) and Bio Numerics software. Results Vibrio parahaemolyticus nucleic acid was detected from all 88 strains. All strains were sensitive to the 8 antibiotics. All strains were divided into 38 PFGE types( PT) and 8 groups( A to H).Group F was the predominant group. Conclusion Combination of real time fluorescence quantitative PCR and the conventional culture method can shorten detection time and raise detection rate. All strains are sensitive to the 8 antibiotics and have close genetic relationship.
