发情期大熊猫阴道源产H2O2乳酸菌的分离鉴定及筛选
Isolation and identification of lactic acid bacteria from the vagina of oestrum giant panda and screening of producing-H2O2 isolates
【摘要】 为了筛选大熊猫阴道中具有产过氧化氢特征的阴道乳酸菌,通过菌落形态特征、生化特性及触酶反应等对10只发情期大熊猫阴道乳酸菌进行分离鉴定,获得42株乳酸菌,经产过氧化氢半定量试验筛选出18株产H2O2的乳酸菌。将18株乳酸菌通过16S r DNA测序并进行NCBI Blast分析,鉴定出粪肠球菌(Enterococcus faecalis)12株、泰国肠球菌(Enterococcus thailandicus)2株、屎肠球菌(Enterococcus faecium)1株、耐久肠球菌(Enterococcus durans)1株、海氏肠球菌(Enterococcus hirae)1株、巴氏链球菌(Streptococcus pasteurianus)1株。用Neighbor-Joining Tree方法构建的系统进化树表明,这18株乳酸菌的16S r DNA以100%的相似性聚在同一个分支上。上述结果为进一步研究大熊猫阴道益生菌奠定了基础。
【Abstract】 In order to screen the vaginal lactic acid bacteria with hydrogen peroxide-producing feature in giant panda, we have been got 42 isolates of lactic acid bacteria from 10 estrus giant pandas’ vagina by colony morphology,biochemical characteristics,and catalase reaction.Meanwhile,18 isolates of lactic acid bacteria with producing H2O2 feature have been got by semi-quantitative test.We have been iden tified 12 isolates of Enterococcus faecalis,2 isolates of Enterococcus thailandicus,1isolates of Enterococcus faecium,1 isolates of Enterococcus durans,1 isolates of Enterococcus hirae,and 1 isolates of Streptococcus pasteurianus by 16 S r DNA sequencing and NCBI Blast analysis.The phylogenetic tree,constructed by Neighbor-Joining method,showed that the 16 S r DNA sequence of 18 isolates of lactic acid bacteria were together on the same branch with 100%similarity.These results laid the foundation for further study of the giant panda vaginal probiotics.
