Metalnikowin类抗菌肽MLP的设计及其抗菌活性的测定
Molecular design and antibacterial activity determination of Metalnikowin-like antibacterial peptide
【摘要】 为设计新型Metalnikowin类抗菌肽并评价其抗菌活性,依据抗菌肽Metalnikowin的作用机理和氨基酸组成,通过人工方法设计合成Metalnikowin类抗菌肽(Metalnikowin-like antibacterial peptide,MLP)并测定其体外和体内抗菌活性。体外抑菌试验结果显示,抗菌肽MLP对鸡致病性大肠杆菌O 1、猪致病性大肠杆菌K 99(A TCC39302)、鸡白痢沙门菌(ATCC10398)、猪霍乱沙门菌(ATCC13312)、禽多杀性巴氏杆菌(CVCC474)和金黄色葡萄球菌(ATCC29213)均具有很好的抑菌活性,其对这6种细菌的最小抑菌浓度分别为1.56、6.25、3.12、6.25、1.56和12.5 g/m L。酸稳定性试验结果显示,抗菌肽MLP在p H值为5~6时活性最高。热稳定性试验结果显示,抗菌肽MLP 100℃加热40 min后仍具有抑菌活性。在对雏鸡感染鸡大肠杆菌和小鼠感染金黄色葡萄球菌的体内疗效试验中,抗菌肽MLP显示了较好的治疗效果。本研究为临床上设计有效的Metalnikowin类抗菌肽提供了参考数据。
【Abstract】 To design and evaluate antibacterial activity of a novel Metalnikowin-like antibacterial peptide(MLP),based on antibacterial mechanism and amino acid composition of antibacterial peptide Metalnikowin,we designed and synthesized an MLP,and determined its biological characteristics.The antibacterial activity assay in vitro showed that the peptide MLP had higher antibacterial activity against chicken enteropathogenic Escherichia coli O1,swine enteropathogenic Escherichia coli K99(ATCC39302),chicken Salmonella pullorum(ATCC10398),Salmonella choleraesuis(ATCC13312),Pasteurella multocida(CVCC474) and Staphylococcus aureus(ATCC29213).The minimal inhibitory concentration of MLP against the six strains were 1.56 g/m L,6.25 g/m L,3.12 g/m L,6.25 g/m L,1.56 g/m L and 12.5g/m L, respectively.Moreover,MLP could keep antibacterial activity after being boiled at least 40 minutes and had higher acid stability.The results of therapeutic tests showed that the antibacterial peptide MLP displayed preferable curative effects and protection against chicken enteropathogenic E.coli infection in chicks and S.aureus infection in mice.This study provides valuable support for further development of antibacterial peptide.
