【摘要】 目的探讨缺氧诱导因子(HIF)-1α蛋白在乳腺癌MCF-7细胞中的分布及其意义。方法采用免疫组化和Western blot技术分别检测乳腺癌MCF-7细胞、细胞浆和细胞核中HIF-1α蛋白的表达情况。结果免疫组化结果显示,HIF-1α阳性表达为棕黄色颗粒,主要分布在细胞浆,少数分布在细胞核;同时Western blot结果显示,MCF-7细胞中表达HIF-1α,细胞浆中表达明显高于细胞核(P<0.01)。结论 HIF-1α蛋白主要分布在乳腺癌细胞浆中,需进入细胞核与HIF-1β结合形成具有活性的HIF-1。更多还原

【Abstract】 Objective To explore the distribution and significance of hypoxia inducible factor-1α( HIF-1α) protein in breast cancer MCF-7 cells.Methods The expressions of HIF-1α protein in cytoplasm and nucleus of breast cancer MCF-7 cells were detected by immunohistochemistry( IHC) and Western blot respectively.Results Immunohistochemical results showed that positive HIF-1α expression was colored brown-yellow particles,which mainly located in cytoplasm,and few located in nucleus.Western blot results showed that the expression level of HIF-1α in cytoplasm of breast cancer MCF-7 cells was statistically significantly higher than that in nucleus( P< 0.01).Conclusion HIF-1α protein mainly locates in cytoplasm of breast cancer cells,and it must enter into nucleus to combine with HIF-1β,then HIF-1 with activity forms.更多还原
