高压氧联合rh-EPO对窒息新生大鼠心肌细胞caspase-3表达的影响

【摘要】 目的探讨高压氧联合rh-EPO对窒息新生大鼠心肌细胞caspase-3表达及细胞凋亡的影响,为临床治疗新生儿窒息后心肌损害提供分子理论依据。方法将新生3~7 d的健康SPF级Wistar大鼠随机分为对照组、窒息组、rh-EPO组、高压氧组、高压氧联合rh-EPO组,每组8只,分别给予相应的干预措施后,采用免疫组织化学染色法检测Caspase-3蛋白表达水平,采用TUNEL染色检测心肌细胞的凋亡率。结果与rh-EPO组和高压氧组比较,高压氧联合rh-EPO组可明显降低窒息新生大鼠心肌细胞caspase-3的表达和细胞凋亡,差异有统计学意义(P<0.05)。结论新生大鼠窒息后采用高压氧及rh EPO联合治疗可明显减轻窒息新生大鼠心肌细胞的损害。

【Abstract】 Objective To explore the effect of hyperbaric oxygen combined with recombinant human erythropoietin(rh-EPO) on Caspase-3 expression and apoptosis of myocardial cells in neonatal rats with asphyxia,provide a molecular basis for clinical treatment of myocardial damage after neonatal asphyxia.Methods Healthy specific- pathogen free Wistar rats aged 3-7 days were randomly divided into control group,asphyxia group,rh- EPO group,hyperbaric oxygen group,hyperbaric oxygen combined with rh- EPO group,8 rats in each group.The rats in different groups were treated by corresponding measures.Immunohistochemical staining was used to detect the expression levels of Caspase- 3 protein.TUNEL staining was used to detect apoptotic rate of myocardial cells.Results Compared with rh- EPO group and hyperbaric oxygen group,the expression level of Caspase-3 protein and apoptotic rate of myocardial cells in hyperbaric oxygen combined with rh- EPO group decreased,there were statistically significant differences(P<0.05).Conclusion Hyperbaric oxygen combined with rh- EPO can significantly relieve damage of myocardial cells in neonatal rats after asphyxia.
