准噶尔乌头中4种生物碱在炮制过程中的含量变化

【摘要】 目的通过HPLC法测定准噶尔乌头Aconitum soongaricum Stapf.中4种生物碱的含有量,并研究其随炮制时间的变化。方法准噶尔乌头乙醚提取液的分析采用XBridge TM C18色谱柱(250 mm×4.6 mm,5μm);以甲醇-水-氯仿-三乙胺(67∶33∶2∶0.1)为流动相,等度洗脱;体积流量0.8 m L/min;检测波长235 nm;柱温40℃。加热水煮法炮制该药材,比较各生物碱含有量随炮制时间(0~255 min)的变化。结果准噶尔乌头碱、12-表-欧乌头碱、乌头碱、脱氧乌头碱平均含有量分别为1.111 3、1.980 2、6.562 6、0.270 9 mg/g。随着炮制时间增加,乌头碱相对峰高变化最显著,脱氧乌头碱逐渐消失,准噶尔乌头碱以和12-表-欧乌头碱较稳定。在18 min和25 min出现了新色谱峰,无明显变化规律。结论准噶尔乌头中上述4种生物碱经过加热水煮炮制后均发生了水解,可达到减毒效果。

【Abstract】 AIM To determine the contents of four alkaloids in Aconitum soongaricum Stapf. by HPLC and to study their changes with processing time. METHODS The analysis of A. soongaricum ether extract was performed on a 40 ℃ thermostastic XBridge TM C18column( 250 mm × 4. 6 mm,5 μm),with the mobile phase comprising of methanol-water-chloroform-triethylamine( 67 ∶ 33 ∶ 2 ∶ 0. 1) flowing at 0. 8 m L / min in an isocratic elution manner,and the detection wavelength was set at 235 nm. The medicinal material was processed with heated water boiling,then the changes of various alkaloids’ contents with processing time( 0 ~ 255 min) were compared.RESULTS The average contents of songorine,12-epi-napellin,aconitine and deoxyaconitine were 1. 111 3,1. 980 2,6. 562 6 and 0. 270 9 mg / g,respectively. With the increase of processing time,the relative peak height of aconitine changed the most significantly,that of deoxyaconitine disappeared gradually,while those of songorine and 12-epi-napellin were relatively stable. At 18 min and 25 min,new chromatographic peaks appeared,which showed no obvious change law. CONCLUSION After processing with heated water boiling,the aformentioned four alkaloids in A. soongaricum are all hydrolyzed,which can achieve attenuated effect.
