一株喜树内生菌的分离与发酵试验

【摘要】 喜树碱是从喜树(Camptotheca acuminata Decne.)里提取的一种抗癌药物。但由于喜树中喜树碱的含量十分低,喜树碱的获取十分困难。在这个研究中,我们采用了组织块法从喜树的果实、树皮、叶片中分离筛选能够生产喜树碱的内生真菌,一共从分离得到了15株菌株。经过摇瓶发酵培养后,用TLC法与HPLC法对其菌丝体提取物进行分析,发现有1株菌株能够产生喜树碱(Camptothecin,CPT),其喜树碱产量为227μg/L。且该菌株在常规的PDA液体培养基中易于培养、生长迅速,表明该菌是一株优良的喜树碱产生菌。

【Abstract】 Camptothecin(CPT) is a kind of anti-cancer drug consisted in Camptotheca acuminata Decne,but it is hard to obtain due to Camptotheca acuminata Decne’s very low content of CPT.In this work,to efficiently obtain CPT,we used tissue block method to isolate and purify endophytes that can produce CPT from fruits,branches and leaves of Camptotheca acuminata Decne.15 endophyte strains were obtained and cultivated in flasks.Then TLC and HPLC methods were used to analyze the extracts of endophyte hypha.The results revealed that one of the 15 strains can produce CPT as many as 227 μg / L.In addition,this strain have high reproducing speed in the PDA medium,indicating it is a very good producer of CPT.
