HPLC测定柴葛清热颗粒中柴胡皂苷a、葛根素和黄芩苷含量
Determination of Saikosaponin a, Paeoniflorin and Baicalin in Chaigeqingre Granula by HPLC
【摘要】 目的建立柴葛清热颗粒中柴胡皂苷a、葛根素和黄芩苷含量的测定方法。方法采用HPLC法,色谱柱为Agilent-C18色谱柱(150 mm×4.6 mm,5μm),流动相:0.1%磷酸水溶液(A)-乙腈(B),梯度洗脱(0~12 min,15%~20%B;12~30 min,20%~25%B),流速为1.0 m L·min-1,检测波长210 nm,柱温30℃,进样量20μL。结果胡皂苷a在1.96~31.36μg·m L-1,葛根素在9.4~150.4μg·m L-1,黄芩苷1.52~24.32μg·m L-1范围内线性关系良好。平均回收率分别为97.65%、103.56%、102.83%,RSD分别为1.29%、1.14%、1.20%。结论该方法准确简便重复性好,可用于柴葛清热颗粒中柴胡皂苷a、葛根素和黄芩苷的含量测定。
【Abstract】 Objective To establish a method for the determination of saikosaponin a, paeoniflorin and baicalin in Chaigeqingre granula by HPLC. Methods The chromatographic column was Agilent-C18(150 mm×4.6 mm, 5 μm), using the gradient mobile phase composed of 0.1% H3PO4(A)-acetonitrile(B),(012 min, 1520% B; 1230 min, 2025% B), the flow rate was 1.0 m L·min-1 and the detection wavelength was 210 nm. The temperature of column was 30 ℃, and the injection volume was 20 μL.Results The linear range of saikosaponin a, paeoniflorin and baicalin was 1.9631.36 μg·m L-1, 9.4150.4 μg·m L-1 and 1.5224.32 μg·m L-1with average recovery of 97.65%(RSD=1.29%), 103.56%(RSD=1.14%), 102.83%(RSD=1.20%). Conclusion The method is convenient, accurate and reproducible for the determination of saikosaponin a, paeoniflorin and baicalin in Chaigeqingre granula.
