The elbow venous blood collected in disposable anticoagulant tube that containing heparin,using RMPI1640 medium and cultured in vitro. Through control dosage of blood,colchicines add time and concentration,hypotonic time,drop height and other conditions,It is concluded that the amount of 12 to 15 drops of blood,the concentration of colchicines for 100 ug / ml,can get good shape,size medium,distributed evenly,contour clear metaphase chromosome specimens.
