乳浆大戟抑制人宫颈癌细胞增殖迁移和诱导细胞凋亡的研究

【摘要】 目的研究乳浆大戟抑制人宫颈癌细胞增殖、迁移、侵袭和诱导细胞凋亡的作用并初步探讨其作用机制。方法用不同稀释度的乳浆大戟提取液处理人宫颈癌Si Ha细胞,MTT实验观察细胞生长抑制现象,吖啶橙染色荧光显微镜观察凋亡细胞的核形态改变并测定凋亡指数,流式细胞术检测细胞凋亡率,Transwell法检测细胞迁移能力和侵袭能力,用分光光度法检测caspase-3和caspase-9的表达情况。结果乳浆大戟对人宫颈癌Si Ha细胞有增殖抑制作用,该作用有时间和浓度依赖性。乳浆大戟处理后的人宫颈癌Si Ha细胞,凋亡指数和凋亡率均显著上升,均有时间和浓度依赖性。Transwell法检测显示,乳浆大戟提取液处理后的Si Ha细胞,迁移能力和侵袭能力显著下降。分光光度法检测发现,乳浆大戟处理后的Si Ha细胞中,caspase-3和caspase-9均表达增强。结论乳浆大戟提取液具有抑制人宫颈癌Si Ha细胞增殖和诱导细胞凋亡的作用,其作用通路有caspase-3和caspase-9的参与。

【Abstract】 Objective To study the effects of Euphorbia esula in inhibition of human cervical cancer cell proliferation,migration,invasion,and apoptosis induction and its preliminarily mechanism. Methods Different dilutions of Euphorbia esula extract were used to treat human cervical cancer Si Ha cells; the cell growth inhibition phenomenon was determined by MTT experimen. Acridine orange staining and fluorescence microscope was used to observe the nuclear morphological changes of apoptotic cells and to determine the apoptotic index. Flow cytometry was used to detect apoptosis rate. Transwell method was used to detect cell migration and invasion ability. Spectrophotometry was used to detect caspase- 3 and caspase- 9 expressions. Results Euphorbia esula had the proliferation inhibition effect on cervical cancer Si Ha cells,which was time and concentration dependent. After Euphorbia esula treatment of cervical cancer Si Ha cells,apoptosis index and apoptosis rate were significantly increased,having the time and the concentration dependence. Transwell method detection showed that the migration and the invasion ability dropped significantly in Euphorbia esula extract processed Si Ha cells. Spectrophotometry showed increased caspase- 3 and caspase- 9 expression in Euphorbia esula processed Si Ha cells. Conclusion The Euphorbia esula extract could inhibit human cervical cancer Si Ha cell proliferation and induce cell apoptosis; the pathway had caspase- 3and caspase- 9 participation.
