apelin-13对小鼠脑出血后脑水肿的影响
Effects of apelin-13 on mice with brain edema after ICH
【摘要】 目的 观察apelin-13对小鼠脑出血后脑水肿的影响,探讨其与脑水肿相关蛋白水通道蛋白4(AQP4)和基质金属蛋白酶9(MMP9)表达的关系。方法 取健康雄性CD-1小鼠,随机分为假手术组(Sham组)、apelin-13组和生理盐水组(NS组)。通过向小鼠纹状体注射胶原酶Ⅳ建立脑出血模型,于脑出血模型建立前5 min,侧脑室给予apelin-13 (0.3μg/g)或等量生理盐水。干湿法检测脑出血1、6、12、24、48 h后的脑组织含水量。Western blot检测脑出血24、48 h后AQP4和MMP9蛋白表达情况。结果 与Sham组相比,NS组脑组织含水量在脑出血后6 h开始显著增加,在24 h达到脑含水量高峰,持续到出血后48 h。而与NS组相比,apelin-13组脑组织水含量在出血后24、48 h显著降低。小鼠脑出血后24、48 h,出血区及出血周边脑组织AQP4蛋白、MMP9蛋白表达均下调。结论 apelin-13能够显著抑制脑出血后脑水肿,其作用机制可能与AQP4和MMP9蛋白表达水平的下调有关。
【Abstract】 Objective To observe the effectd of apelin-13 on mice with brain edema after intracerebral hemorrhage(ICH), and to explore the relationship between apelin-13 and the expression of aquaporin-4(AQP4), matrix metalloproteinase-9(MMP9). Methods Healthy male CD-1 mice were randomly divided into three groups: a sham-operated group, a normal saline(NS) group and an apelin-13 group. A mice model of ICH was established by injection of collagenase IV to the striatum. Then, 5 min before ICH 0.3 g/g apelin-13 or the same volume of normal saline was intracerebroventricularly administered. The cerebral water content(CWC) was measured by the dry-wet method, while the amounts of AQP4 and MMP-9 were determined by Western blotting. Results Compared with the sham-operated group, the CWC stated to significantly increase in Group NS 6 h after ICH and continued for 48 h, reaching a peak 24 h after ICH. Compared with Group NS, the CWC was remarkably decreased in Group apelin-13 24 h and 48 h after ICH. Meanwhile, the amounts of AQP4 and MMP9 were down regulated by apelin-13 in the ICH area and its surrounding tissues. Conclusion Apelin-13 can attenuate brain edema after ICH in mice, which may be associated with the down-regulation of AQP4 and MMP9 protein expression.
