复方苦参凝胶对豚鼠实验性急性湿疹的作用及机制

【摘要】 目的探讨复方苦参凝胶对豚鼠急性湿疹模型的治疗作用及作用机制。方法健康豚鼠60只随机分为6组,每组10只。除空白对照组外其余各组均采用反复涂抹2,4-二硝基氯苯(DNCB)建造豚鼠急性湿疹模型。造模成功后,空白对照组,模型组,苦参凝胶低、中、高剂量组和阳性对照组豚鼠分别给予空白凝胶,空白凝胶,含苦参碱25、50、100 mg·g-1的复方苦参凝胶和糠酸莫米松凝胶干预,剂量均为0.5 g·kg-1,bid。实验第22日观察各组豚鼠右背部皮肤病变并评分,测量皮肤增厚及增重程度。ELISA法测定皮肤组织中TNF-α及IL-1β水平,Western blot法测定皮肤组织中ICAM-1及VCAM-1的表达水平。结果与空白对照组比较,模型组豚鼠皮肤出现肿胀、红斑、糜烂和渗出等症状,皮肤病变评分增高(P<0.01),皮肤明显增厚、增重(P<0.01);苦参凝胶中、高剂量组及阳性对照组上述指标与模型组比较均有不同程度减轻。与空白对照组比较,模型组豚鼠皮肤组织TNF-α及IL-1β含量增加(P<0.01),ICAM-1及VCAM-1的表达增加(P<0.01);与模型组比较,苦参凝胶中、高剂量组及阳性对照组TNF-α及IL-1β含量降低(P<0.01),ICAM-1及VCAM-1的表达下调(P<0.01)。结论复方苦参凝胶对于豚鼠急性湿疹具有明显的疗效,其机制可能与其调节炎症因子、调控相关细胞黏附分子的表达有关。

【Abstract】 AIM To investigate the effects and mechanism of Fufang Kushen gel on atopic eczema model of guinea pigs. METHODS Sixty healthy guinea pigs were randomly divided into 6 groups on average: normal control group, model group, Kushen gel low dose group, Kushen gel media dose group, Kushen gel high dose group and positive group. The 2, 4- dinitrochlorobenzene smearing was used to make atopic eczema model except the normal control group. After the model established, each guinea pig of six group was treated with placebo-gel, placebo- gel, low dose Kushen gel(contained oxymatrine 25 mg·g-1), media dose Kushen gel(contained oxymatrine 50 mg·g-1), high dose Kushen gel(contained oxymatrine 100 mg·g-1) and mometasone furoate gel0.5 g·kg-1, bid, respectively. Twenty- two days later, all animals were sacrificed. The score of pathological change was detected. The swelling grade was reflected by the weight and thickness of right side skin compared with left side. The content of TNF- α and IL- 1β in skin were detected by ELISA. The express of ICAM- 1 and VCAM- 1 were measured by Western blot. RESULTS Compared with the normal control group, some syndromes such as swelling, erythema, erosion and effusion appeared in the model group, and the score and the swelling grade of model group were sharpen and heighten(P < 0.01). The content of TNF- α and IL- 1β and the express of ICAM- 1 and VCAM- 1 in the model group were increased compared to the normal control group(P < 0.01).Compared with the model group, the skin pathological change were ameliorated and the score and swelling degree were lowered in the Kushen gel media, high dose group and positive group( P < 0.01), while the content of TNF- α and IL- 1β and the express of ICAM- 1 and VCAM- 1 were decreased( P < 0.01).CONCLUSION Fufang Kushen gel has therapeutic effects on atopic eczema in guinea pigs, and its mechanism maybe related to reducing the content of TNF- α and IL- 1β and the express of ICAM- 1 and VCAM- 1.
