苯丁酸氮芥抗套细胞淋巴瘤细胞株Jeko-1作用机制的研究

【摘要】 目的:探讨苯丁酸氮芥抗套细胞淋巴瘤细胞株Jeko-1作用机制。方法:应用MTT法检测苯丁酸氮芥对Jeko-1细胞增殖的影响,Hoechst染色及Annexin V-FITC双染检测苯丁酸氮芥对Jeko-1细胞凋亡的影响,Western blot检测苯丁酸氮芥对Jeko-1细胞凋亡相关蛋白BAX,BCL-2,procaspase 3,procaspase 8,procaspase 9表达水平及PI3K/AKT信号通路的激活情况。结果:苯丁酸氮芥0、5、10和20μmol/L作用Jeko-1细胞24、48、72 h后,细胞增殖受到抑制,并呈时间及剂量依赖性(r=0.873,r=0.932)。0,5,10,20μmol/L苯丁酸氮芥作用Jeko-1细胞72 h后,细胞凋亡率显著提高,BAX,procaspase 3,procaspase 8及procaspase 9蛋白表达水平显著上调,BCL-2表达水平显著下调,PI3K蛋白及AKT磷酸化水平显著降低。结论:苯丁酸氮芥能显著诱导套细胞淋巴瘤细胞株Jeko-1凋亡,其作用机制与调节PI3K/AKT信号通路有关。

【Abstract】 Objective:To explore the action mechanism of chlorambucil against mantle cell lymphoma cell line Jeko-1.Methods:The effect of chlorambucil on Jeko-1 cell proliferation was measured by MTT method.The effect of chlorambucil on the apoptosis of Jeko-l cell was detected by Hoechst staining and Annexin V-FITC dual staining.The activation of PI3K/AKT signaling pathway and the expression of BAX,BCL-2,procaspase 3,procaspase 8 and procaspase 9 were detected by Western blot.Results:0,5,10,20 μmol/L chlorambucil could inhibit Jeko-l cell proliferation at 24,48,72 h in a time-and dose-dependent manner.Chlorambucil of 0,5,10,20 μmol/L increased the apoptotic rate of Jeko-1 cells,upregulated the expression of BAX,procaspase 3,procaspase 8,procaspase 9 and PI3 K,increased the phosphorylation of AKT and down-regulated the expression of BCL-2.Conclusion:The chlorambucil can induce the apoptosis of mantle cell lymphoma Jeko-l cells via blocking PI3K/AKT signaling pathway.
