一株猪轮状病毒的分离与鉴定

【摘要】 将1份经RT-PCR检测猪轮状病毒阳性的临床腹泻粪样感染Vero细胞,连续盲传8代,出现病变,成功分离到1株病毒,经RT-PCR检验和电镜观察,鉴定该病毒为轮状病毒,命名为GD-01-2015。扩增该病毒的VP4和VP7基因进行分型和系统进化分析,结果发现,其VP4基因为P[7]型,与来自韩国的猪源毒株K71同源性达到99.8%;其VP7基因为G5型,与来自韩国的猪源毒株06-6-1同源性达到99.7%。由此可以推测GD-01-2015株与韩国猪源毒株有相似的进化来源。根据轮状病毒分类委员会(RCWG)提出的A群轮状病毒的最新分类方法,GD-01-2015株基因型为G5P[7]。本研究为监测轮状病毒的流行状况及其疫苗的研制提供了理论依据。

【Abstract】 One viral strain,GD- 01- 2015,was successfully isolated from fecal samples from a diarrheic piglet,which was identified as porcine rotavirus infection by RT- PCR. This strain was passaged eight times in a monolayer of Vero including the initial adaptation,and identified to be porcine rotavirus by RT- PCR and electron microscopic observation. The nucleotide sequences of VP4 and VP7 were obtained and compared with those of the other known porcine group A rotavirus( RVA) strains. According to the phylogenetic and molecular evolutionary results,VP4 gene of the isolated strain was shown to possess a typical porcine- like genotype constellation similar to that of the P[7]prototype and shared 99. 8% nucleotide identities with the porcine RVA strain K71 isolated from South Korea; VP7 gene was similar to G5 prototype and shared 99. 7% with the porcine RVA strain 06- 6- 1,which was also isolated from South Korea. It suggests that this strain has a strong connection with those strains from South Korea and,according to the genotyping recommendations of the Rotavirus Classification Working Group( RCWG),this strain should be classified as G5 P [7]prototype. This study provides a basis for investigation of RVA epidemic status and vaccine development.
