【摘要】 目的研究异常黏液质型阳痿病证大鼠生精细胞凋亡及维药伊木萨克片对其影响。方法选用100只雄性SD大鼠,随机选取10只为正常组,余90只为造模组,以湿寒饲料+湿寒性环境的干预条件建立异常黏液质证候模型,20w后从证候组中筛选出阳痿病证模型后,将其分为病证模型组和病证药物干预组,剩余证候模型分为证候模型组、证候药物干预组,干预2w,取材后对睾丸用苏木精-伊红(hematoxylin-eosin,HE)染色后光镜下观察其睾丸组织病理改变,原位末端转移酶标记技术(TdT-mediated dUTP Nick-End Labeling,TUNEL)法检测生精细胞的凋亡情况。结果 (1)HE染色结果:正常组结构完好;证候模型组生精细胞数量明显少于正常组,间质细胞及精子数量减少;病证模型组生精小管萎缩或扭曲成不规则形,生精细胞和间质细胞数量显著少于正常组;证候药物干预组生精小管形态、生精细胞数量及管腔内精子数量基本同正常组,间质细胞增多;病证药物干预组生精细胞数量较病证模型组有所增多,精子数量依旧少于正常组。(2)凋亡检测结果:证候模型组及病证模型组生精细胞凋亡较正常组显著增多(P<0.01),经伊木萨克片干预后凋亡数量下降(P<0.01)。结论维医异常黏液质型阳痿病证大鼠模型睾丸组织发生病理学改变,生精细胞发生异常凋亡,伊木萨克片干预可有效减少异常凋亡。更多还原

【Abstract】 Objective To study the apoptosis of germ cell of abnormal phlegmatic impotence rat’s testicular tissue and the interventional influence using Yimsake.Methods A total of 100 normal mature male SD rats were enrolled in the study,10 of them were randomly chosen as the nomal group,the other 90 were treated with spinach and coriander diet in a cold and humid until the model of abnormal phlegmatic syndrome was established.Biological characteristics of rats were observed about 20 weeks until establishment abnormal phlegmatic group,then through APO erectile experiment and matching experiment,the rats were randomly divided into abnormal phlegmatic syndrome group,medication intervention of abnormal phlegmatic syndrome group,abnormal phlegmatic syndrome impotence disease model group,and medication intervention group of abnormal phlegmatic syndrome impotence group.After2 weeks intervention,hematoxylin-eosin(HE)staining was carried out to observe the morphological changes of all rats’ s testicular tissue and then through TdT-mediated dUTP Nick-End Labeling to test germ cell apoptosis.Results HE staining result showed that the morphological structure of normal group is normal.Compared with normal group,the quantity of abnormal phlegmatic group’s germ cells was significantly decreased and its leydig cells and sperm were decreased as well.Compared with normal group the testicular morphological structure of abnormal phlegmatic impotence group was twisted and out of order and its germ cells were significantly fewer.The morphological structure and quantity of germ cells of medical intervention abnormal phlegmatic group was almost the same as normal group.Compared with abnormal phlegmatic impotence group the quantity of germ cell of medical intervention abnormal phlegmatic impotence group was increased but still less than normal group.(2)Apoptosis detection result showed that the abnormal phlegmatic syndrome group and the abnormal phlegmatic impotence group germ cells apoptosis was significantly increased compared with normal group(P <0.01)and after Yimusake tablet intervention,both model groups germ cells apoptosis level was markedly decreased(P <0.01).Conclusion The testicular morphological structure of Uyghur medical abnormal phlegmatic with impotence rats was histopathologically changed,germ cells had apoptosis,those pathological situations recovered after Yimusake tablet medication intervention.更多还原
