红花注射液对缺血缺氧性损伤的兔海马CA1神经元nNOS表达的影响

【摘要】 目的研究红花注射液(SY)对控制性降压(CH)诱导的缺血缺氧性损伤兔海马CA1神经元nNOS表达的影响。方法构建缺血缺氧性脑损伤动物模型,SY组在降压前30min静脉注射红花注射液2mL/kg(1mL含SY1.6mg)。nNOS免疫组化染色观察CA1神经元形态学改变,Westernblotting检测nNOS蛋白的表达水平。结果SY组nNOS免疫组化染色的光密度值显著低于CH组(P<0.05),其nNOS蛋白表达水平也明显低于CH组,但高于Normal组(P<0.05)。结论红花注射液显著减轻CH诱导的兔海马缺血缺氧性损伤,可能是通过抑制nNOS蛋白表达起作用的。

【Abstract】 Objective To study the effect of safflower injection( SY) on n NOS expression of hippocampal CA1 neurons suffering from hypoxic- ischemic injury induced by controlled hypotension( CH) in rabbits. Methods The hypoxic-ischemic brain injury model was constructed and the SY group was intravenously injected with 2 mL / kg of safflower injection( 1 m L,containing 1. 6 mg SY) 30 min before CH. The neuronal changes in hippocampal CA1 were observed for n NOS immunohistochemical staining,and western blotting was used to detect the expression of n NOS. Results Optical density values of n NOS staining in the SY group were significantly lower than that in the CH group( P < 0. 05). The expression of n NOS in the SY group was significantly lower than that in the CH group,but it was higher than that in the normal control group( P <0. 05). Conclusion SY can significantly attenuate the hypoxic- ischemic hippocampal injury,possibly by inhibiting n NOS expression.
