饮食干预对妊娠期糖尿病小鼠子代中远期瘦素及糖脂代谢的影响
Long-term effect of diet intervention on leptin,glucose and lipid metabolism in the offspring of gestational diabetes mellitus mice
【摘要】 目的研究妊娠期糖尿病(GDM)子代小鼠在饮食干预下其瘦素水平及糖脂代谢的转归情况。方法链脲霉素腹腔注射建立GDM模型,将其产下子鼠随机分为对照组、高脂组(G-HF)、高纤组(G-HX)、热量限制组(G-CR),非GDM子鼠建立空白对照组(C-CON);每组各7只。干预后测总胆固醇(TCH),甘油三酯(TG),低密度脂蛋白胆固醇(LDL-C),高密度脂蛋白胆固醇(HDL-C),胰岛素、瘦素水平。结果 (1)GDM子代血糖显著高于正常鼠子代(P<0.05);TCH,TG,LDL-C显著升高(P<0.05)。(2)饮食干预后,热量限制组血糖体重趋于稳定;高脂组血脂代谢水平均显著降低(P<0.05);高纤组HDL-C高于空白对照组(P<0.05);热量限制组TCH,TG,LDL-C明显降低(P<0.001),瘦素、胰岛素改善。(3)血清瘦素水平与TG、TCH、LDL-C及胰岛素水平正相关(P<0.05)。结论 GDM子鼠有明显的不良结局。高脂饮食能加重GDM子代不良转归,高纤饮食作用较弱,热量限制可能通过改善瘦素抵抗,显著改善胰岛素抵抗及糖脂代谢。
【Abstract】 Objective To study the long-term effect of diet intervention on the offspring of gestational diabetes mellitus( GDM) mice in leptin,glucose,and lipid. Methods GDM pregnant mice pattern was built by injecting streptozotocin( STZ),and the offspring mice were randomly divided into four groups: control group( G-CON),high-fat diet Group( G-HF),high fiber diet Group( G-HX) and calorie restriction Group( G-CR). The offspring in non-GDM pregnant mice were as the blank control group( C-CON). Each group included 7 offspring mice. After the experiment,we measured the total cholesterol( TCH),triglycerides( TG),low-density lipoprotein cholesterol( LDL-C),high-density lipoprotein cholesterol( HDL-C),insulin and leptin levels. Results( 1) The blood glucose in GDM descendants was significantly higher than that in normal offspring( P < 0. 05). Meanwhile,TCH,TG and LDL-C increased significantly( P < 0. 05).( 2) After the dietary intervention,the weight of calorie-restricted group became stable,while the lipid metabolism levels of the high-fat group were significantly lower( P < 0. 05). The HDL-C in high fiber group was higher than that in the control group( P < 0. 05). The TCH,TG,LDL-C of the calorie-restricted group decreased significantly( P < 0. 001),and the levels of leptin and insulin improved.( 3) The levels of leptin were a positive correlation with TG,TCH,LDL-C and insulin levels( P < 0. 05).Conclusion GDM has the obvious adverse outcomes in offspring of GDM mice. High-fat diet could aggravate the adverse outcomes of GDM offspring. The effects of high fiber diet are weaker. At the same time,calorie restriction may improve insulin resistance,leptin resistance,and glucose and lipid metabolism.
