甘草对去势大鼠骨质疏松症骨形态计量学影响的研究

【摘要】 目的观察甘草对去势大鼠骨质疏松症模型骨组织结构的影响,探讨其干预骨质疏松的作用机制,为临床研究提供实验依据。方法建立去势大鼠骨质疏松模型,将造模成功的45只雌性SD大鼠按随机数字法分为去势组、甘草汤低剂量组、甘草汤高剂量组,每组15只。去势组给予生理盐水灌胃,甘草汤低剂量组和甘草汤高剂量组分别给予甘草汤剂2 m L和4 m L灌胃,每周1次,连续16周。末次灌胃后24 h取大鼠胫骨组织,制作冰冻切片冯库萨(von kossa)钙染色观察骨组织形态结构,采用免疫组化和图像分析技术测定骨小梁结构的表达情况。结果去势组仅骨小梁结构少量表达;甘草汤低剂量组骨小梁组织表达相对增强,骨小梁数量增多、相对体积增大、分离度减小;甘草汤高剂量组骨小梁组织表达明显增强,骨小梁数量、相对体积进一步增大而分离度显著减小。甘草汤高、低剂量组骨小梁组织阳性目标总面积、骨小梁总周长及骨小梁数量均明显高于去势组(P均<0.05),骨小梁分离度明显低于去势组(P均<0.05)。结论甘草能够增加成骨细胞中骨小梁的数量而保护骨组织,增加骨量,并能防止骨基质的丢失,从而增加骨的韧性,可用于绝经后骨质疏松症的治疗。

【Abstract】 Objective It is to investigate the effects of glycyrrhiza on the bone histomorphometry in ovariectomized osteoporotic rats,and explore the intervention mechanism of osteoporosis,so as to provide experimental basis for clinical research.Methods The ovariectomized rat models of osteoporosis were established,45 SD female rats were randomly divided into 3groups,ovariectomized control group( A),short dose of glycyrrhiza group( B),high dose of glycyrrhiza group( C),15 rats in each group. By corresponding method of intervention,the hismorphological structure of bone tissue sections were observed after staining by von kossa calcium staining,and the bone trabecula expression was detected by immunohistochemistry and image analysis technique. Results There was a little expression of trabecular bone structure in the ovariectomized control group( A); and the expression of the trabecular bone tissue,trabecula bone number,relative volume were increased,the degree of separation decreased in the short dose of glycyrrhiza group( B); and the expression of the trabecular bone tissue,trabecula bone number,relative volume were further increased and the degree of separation significantly decreased in high dose of glycyrrhiza group( C). Compared with the ovariectomized control group( A),the trabecular bone area,trabecular bone perimeter,trabecular bone number,trabecular bone degree of separation all have a statistically significant difference( P < 0. 05). Conclusion Glycyrrhiza can protect bone tissue through raise bone cells and the bone trabecular number,prevent the loss of bone matrix,increasing the toughness of bone. Consequently glycyrrhiza has an distinct curative effect on osteoporosis.
