Lin28a表型小鼠乳腺细胞体外增殖的初步研究
In vitro proliferation of Lin28a phenotype mice mammary cells
【摘要】 目的:研究Lin28a表型小鼠乳腺细胞在体外培养增殖的相关特性。方法:提取Lin28a表型和野生型小鼠乳腺细胞,进行体外传代培养,记录生长曲线,比较传代间的变化,收集细胞,提取RNA和蛋白,检测与DNA甲基化相关基因的变化。结果:1将Lin28a过表达8周未育小鼠乳腺细胞和野生型比较,前者增殖较明显;2P16随传代表达增强,但Lin28a表型使其在传代早期受到更多的抑制;3EZH2,SUZ12作为DNA组蛋白甲基化成分,在lin28a表型乳腺细胞表达更强。结论:Lin28a小鼠乳腺导管过度发育可能与DNA甲基化组分参与有关,后者与P16等基因构成"开关",调节乳腺导管的发育程度。
【Abstract】 Objective:To study the in vitro proliferation properties of Lin28 aphenotype mice mammary cells.Methods:Cells from Lin28 aand wild type mice were extracted and cultured in vitro,the proliferation curve was recorded and changes between generations were compared.Moreover,RNA and protein were obtained from collected cells to detect corresponding DNA methylation of related genes.Results:It was noticed that mammary cells of 8week virgin Lin28 amice proliferated more notably than that in wild mice.P16 expression was enhanced by subculture,but it was inhibited at early subculture in Lin28 aphenotype.EZH2and SUZ12,as components of DNA histone methylation,were expressed more predominantly in Lin28 acells.Conclusion:DNA histone methylation components might participate in the mammary duct hypergenesis of Lin28 aphenotype mice,which may regulate the mammary duct growth together with P16 expression in a switch manner.
