AP-2α对肺腺癌A549细胞MnSOD表达的作用研究
Study on the role of AP-2α in the expression of Mn SOD in the lung cancer A549 cells
【摘要】 目的:探讨转录因子激活蛋白-2α(activator protein-2 alpha,AP-2α)对锰超氧化物歧化酶(manganese superoxide dismutase,Mn SOD)m RNA和蛋白表达的影响,探讨AP-2α下调Mn SOD表达的可能的分子机制。方法:转染Ad-AP-2α和AP-2αsi RNA进入A549细胞,上调和下调AP-2α后,采用实时定量RTPCR及Western blot检测A549细胞内Mn SOD的m RNA及蛋白质的表达水平。结果:1)Ad-AP-2α转染A549细胞后,Mn SOD m RNA和蛋白质表达均下降;2)AP-2αsi RNA转染A549细胞后,Mn SOD m RNA及蛋白质表达均增加。结论:AP-2α对肺腺癌A549细胞Mn SOD的m RNA和蛋白表达起下调作用。
【Abstract】 Objective: To investigate the effect of AP-2α on Mn SOD gene/protein expression in lung cancer(A549) cells and its possible molecular mechanism of downregulation. Methods: A549 cells were transfected with adenovirus vector overexpressing AP-2α and AP-2α si RNA knockdowning AP-2α. The levels of AP-2α on Mn SOD gene/protein expression were detected by RT-PCR and western blot. Results: 1) After transfected with Ad-AP-2α in A549 cells, the levels of Mn SOD m RNA and protein expression both decreased; 2) the expression of m RNA and protein of Mn SOD increased after AP-2α si RNA transfected in A549 cells. Conclusion: The results revealed that AP-2α can effectively down-regulate Mn SOD gene/protein expression.
