经口灌胃卵清蛋白对哺乳期BN大鼠致敏性的影响

【摘要】 目的使用卵清蛋白经口灌胃致敏不同周龄的BN大鼠,初步探讨哺乳期暴露致敏蛋白对BN大鼠致敏性影响,并了解血清中细胞因子变化。方法选2周龄、4周龄和8周的BN大鼠,经口灌胃1 mg/kg·bw·d剂量卵清蛋白,连续35 d。第28天和第35天时内眦静脉采血,采用酶联免疫吸附法测定血清中OVA特异性Ig E(s Ig E)抗体、细胞因子IL-4、IL-10和IFN-γ浓度。结果第28天时,8周龄组BN大鼠血清中OVA s Ig E浓度显著高于对照组(P<0.05),2周龄和4周龄组与对照组之间差异无统计学意义(P>0.05)。第35天时,4周龄组和8周龄组大鼠血清中OVA s Ig E浓度都显著高于对照组(P<0.05),2周龄组与对照组比较差异无统计学意义。同时第35天细胞因子检测结果,2周龄组BN大鼠血清中IL-10和IFN-γ浓度降低(P<0.01),IL-4浓度变化不大。4周龄组和8周龄组BN大鼠血清中IL-4和IL-10浓度增加(P<0.05),血清中IFN-γ浓度变化不大。结论哺乳期会抑制卵清蛋白致敏BN大鼠,这可能受到细胞因子影响。

【Abstract】 Objective To investigate the effect of immune response after oral ovalbumin(OVA) exposure during maternal milk feeding on Brown Norway(BN) rat pups as well as cytokines. Methods Different age rats(2,4 and 8 weeks) were used to treat with1 mg/d of OVA by gavage for 35 days. The levels of OVA specific serum Ig E,IL-4,IL-10 and IFN-γ were analyzed on the 28 thday and 35 th day by enzyme linked immunosorbent assay(ELISA). Results On day 28,8 week group rat produced significantly higher concentration of OVA s Ig E compared to the control group(P < 0. 05). On day 35,the concentrations of OVA s Ig E in 4 week group and 8 week group were significantly higher than control group(P < 0. 05). In addition,The treatment of OVA significantly reduced the concentrations of IL-10 and IFN-γ in 2 week group(P < 0. 01),and increased the concentrations of IL-4 and IL-10 in 4 week and 8 week group on day 35. Conclusion The oral OVA exposure could restrain the development of food sensitization during maternal milk feeding in BN rats,which may be related to cytokines.
