镧对大鼠星形胶质细胞GRP78、IRE1、JNK、Caspase-3和cPARP表达的影响
Influences of Lanthanum to the expression of GRP78,IRE1,JNK,Caspase-3 and cPARP in the astrocytes of rats
【摘要】 目的探讨镧(lanthanum,La)对大鼠大脑皮质星形胶质细胞(astrocyte,AS)的损害效应及对GRP-78、IRE1、JNK、Caspase-3和c PARP表达的影响。方法 0、0.25、0.5和1.0 mmol/L La Cl3处理大鼠大脑皮质AS细胞24 h,然后用乳酸脱氢酶(lactic dehydrogenase,LDH)检测试剂盒测AS细胞LDH释放量,流式细胞仪测AS细胞凋亡情况,Western blot法测AS细胞GRP-78、IRE1、磷酸化的IRE1(phosphorylated IRE1,p IRE1)、JNK、磷酸化的JNK(phosphorylated JNK,p JNK)、Caspase-3和c PARP表达。结果 0.25、0.5和1.0 mmol/L La Cl3处理的大鼠AS细胞LDH释放量和细胞凋亡率显著高于对照AS细胞,且有剂量-反应关系。0.25、0.5和1.0 mmol/L La Cl3处理的大鼠AS细胞GRP-78、p IRE1、p JNK、Caspase-3和c PARP表达显著高于对照AS细胞,且随La Cl3处理AS细胞的剂量增加而升高,其中1.0 mmol/L La Cl3处理的大鼠AS细胞GRP-78、p IRE1、p JNK、Caspase-3和c PARP表达分别为对照AS细胞的2.60倍、2.96倍、3.93倍、3.38倍和2.93倍。结论 La对大鼠大脑皮质AS细胞的损害效应可能和GRP-78水平升高、IRE1和JNK磷酸化增加及Caspase-3、c PARP表达增强而致凋亡增加有关。
【Abstract】 Objective To investigate the harmful effects and influences to the expression of GRP-78,IRE1,JNK,Caspase-3 and c PARP in the rats’ astrocytes( AS) of cerebral cortex caused by lanthanum( La). Methods AS cells of cerebral cortex in rats were incubated with 0,0. 25,0. 5 and 1. 0 mmol / L La Cl3 for 24 h. The release amount of lactic dehydrogenase( LDH) of AS cells was measured with LDH assay kit,and the apoptosis of AS cells was detected with flow cytometry. The expression levels of glucose regulated protein 78( GRP-78),IRE1,phosphorylated IRE1( p IRE1),JNK,phosphorylated JNK( p JNK),Caspase-3 and c PARP of AS cells were measured with Western blot. Results The release amounts of LDH and apoptotic rates of 0. 25,0. 5 and 1. 0 mmol / L La Cl3-treated AS cells in the cerebral cortex of rats were higher than those of control AS cells significantly,and there were dose-response relationship. GRP-78,p IRE1,p JNK,Caspase-3 and c PARP expression of 0. 25,0. 5 and 1. 0 mmol / L La Cl3-treated AS cells in the cerebral cortex of rats were higher than those of control AS cells significantly,and increased as La Cl3-treated dose increasing. Besides,GRP-78,p IRE1,p JNK,Caspase-3 and c PARP expression of 1. 0 mmol / L La Cl3-treated AS cells in the cerebral cortex of rats were 2. 60,2. 96,3. 93,3. 38 and 2. 93 folds of control AS cells. Conclusion La exerted harmful effects on AS cells,which might be connected with increases of apoptosis induced by La by the means of elevating GRP-78 expression,IRE1 and JNK phosphorylation and Caspase-3,c PARP expression in the AS cells of cerebral cortex of rats.
