山豆根苷及苷元的提取和纯化

【摘要】 目的分析山豆根苷及苷元的提取和纯化的方法。方法采用乙醇提取和硅胶柱层析的方法从山豆根中分离和纯化得到有效成分山豆根苷和山豆根苷元。通过L9(43)正交实验发现山豆根苷提取的最佳条件为:乙醇浓度90%,回流3次,回流时间1h,加10倍溶剂量。使用硅胶柱层析分离方法纯化山豆根苷及苷元,依次使用二氯甲烷:甲醇=20:1,二氯甲烷:乙酸乙酯:甲醇=4:4:2,二氯甲烷:乙酸乙酯=5:5梯度洗脱,最后得到山豆根苷和山豆根苷元的白色粉末。结果经检测,两者的纯度分别为93.1%和94.7%。结论由于山豆根苷和苷元极性较大,需要用高极性溶剂提取,采用乙醇回流提取山豆根的方法简单方便,能最大程度的提取出山豆根中的苷,水解后的苷元和苷的混合物采用硅胶柱色谱分离,分离效果良好,适合于山豆根苷及苷元的提取和纯化。

【Abstract】 Objective To analyze the method of purification and extraction of euchresta japonica glycoside and aglucon.Methods The euchresta japonica glycoside and euchresta japonica aglucon who were the active component were separated and purified from euchresta japonica by using the method of ethanol extraction and silica gel column chromatography. Through L9(43) orthogonal experiment to find the best conditions for the extraction of Sophora tonkinensis glycosides: ethanol concentration 90%, 3 times of reflux, reflux time 1H, plus 10 times the dose of solution. The euchresta japonica glycoside and aglucon were purified by using the Silica gel column chromatography separation method.Using in turn the ratio of 20:1 of dichloromethane and methanol, the ratio of 4:4:2 of dichloromethane, ethyl acetate and methanol, the ratio of 5:5 of methylene chloride and ethyl acetate in the gradient elution, finally obtained the white powder of euchresta japonica glycoside and euchresta japonica glycoside aglucon. Results The purity was 93.1% and 94.7% respectively. Conclusion Because euchresta japonica glycoside and aglucon are more polar, need to be extracted with high polar solvent, using ethanol reflux extraction method of euchresta japonica is simple and convenient, can maximize the extraction of glycoside from euchresta japonica.The mixture of aglucon and glycoside hydrolyzed were separated,which can obstain good separation effect, and is suitable for the purification and extraction of euchresta japonica glycoside and aglucon.
