脑干中5-羟色胺的表达对诊断原发性脑干损伤的意义
Brainstem Serotonin Expression Significance for The Diagnosis of Primary Brain Stem Injury
【摘要】 目的探索5-TH在大鼠脑干损伤模型中不同时间段表达水平的不同及其对脑干损伤的诊断意义和恢复评估。方法采用Marmarou自由落体打击装置对实验组雌性SD大鼠复制脑干损伤模型,于伤后6h、12h、24h、3d、7d取脑干组织,经HE染色和免疫组化等技术研究脑干损伤后脑干中5-HT的动态变化规律。并通过5-HT的表达,研究5-HT表达与脑干损伤后恢复的关系。结果实验组可见典型脑干损伤表现,其病理改变为脑血管收缩与扩张,脑干水肿,细胞周围间隙增宽,脑干实质散在点状出血,微血管充血、管周水肿。5-HT于伤后3h表达开始增强,并在6h达到高峰,在12h之后开始逐渐减少直至恢复。阳性染色见于脑干中缝核群和周围神经网状结构中。结论脑干损伤后病理改变有5-HT的参与,且5-HT的异常表达可以对脑干损伤后的恢复及预后有一定影响,对今后的研究方向有一定指导意义。
【Abstract】 Objective To explore the diagnostic significance and recovery evaluating of 5-TH in rat brain stem injury model for brain stem injury in different levels of different time periods. Methods Marmarou freefall combat means experimental groups of female SD rats copy brainstem injury model;research the dynamic changes of the brainstem of 5-HT after injury 6h, 12 h, 24 h, 3d, 7d, by HE staining and immunohistochemical techniques brain stem injury. And through the expression of 5-HT, 5-HT study the expression of the relationship between the brain stem after injury recovery. Results The visible manifestation of typical brain stem injury, the pathological changes of cerebral vascular contraction and expansion, brainstem edema, widened the gap surrounding cells, brain stem substance scattered spotting, capillary congestion, perivascular edema. 5-HT 3h expression began to increase after injury and peaked at 6h, 12 h after gradually reduced until recovery. Positive staining found in the brainstem raphe nuclei and peripheral nerve network structure. Conclusion The pathological changes in the brain stem injury in the participation of 5-HT and 5-HT abnormal expression may have some impact on recovery and prognosis of brain stem injury, the direction of future research and clinical work has a certain significance.
