Clostridium acetobutylicum的亚硝基胍和钴-60复合诱变
COMPOUND MUTATION OF Clostridium acetobutylicum WITH NITROSOGUANIDINE AND COBALT-60
【摘要】 利用化学诱变剂亚硝基胍及物理诱变剂钴-60对丙酮丁醇梭菌YBD(Clostridium acetobutylicum YBD)进行复合诱变,结合多次传代培养,筛选获得一株编号为500-1的突变株。该菌株经6代培养后丁醇产量为15.40 g/L;且能耐受15 g/L的丁醇;在10%玉米醪培养基中,丁醇产量为15.57 g/L,较出发菌株提高27.62%;在10 L发酵罐发酵实验中,以玉米粉为发酵原料,36 h时突变株丁醇产量可达14.24 g/L,生产速率为0.40 g/(L·h),较出发菌株的生产速率提高207.69%。
【Abstract】 Clostridium acetobutylicum YBD was mutated by chemical mutagens nitrosoguanidine(NTG)and physicalmutagens cobalt-60,and a mutant strain,recorded 500-1 with multiple generations culture was acquired.After sixgenerations culture,the mutation strain could produce butanol of 15.40 g/L and tolerate butanol of 15 g/L.The mutantstrain could produce the butanol quantity reached 15.57 g/L.Compared with the original strain,the butanol quantityincreased by 27.62%,respectively.In the 10 L-fermentation experiment with corn flour as the raw material,the butanolquantity of mutant strain of 14.24 g/L was achieved after 36 hours,and the yield was 0.40 g/(L·h)which was increasedby 207.69% than the yield of initial strain.
