链霉菌BAF-0711产生的抗肿瘤活性成分的分离纯化和结构鉴定

【摘要】 探讨分离、纯化和鉴定链霉菌BAF-0711发酵液中的抗肿瘤活性成分的方法。采用大孔吸附柱层析、硅胶柱层析等方法对该菌株的次级代谢产物进行分离纯化;根据理化性质和波谱学方法进行化学结构的鉴定;利用MTT法来检测化合物的抗肿瘤活性。从该菌株发酵液中分离到化合物BAF-0711-A。经波谱方法鉴定与文献中报道的bafilomycin A1同质。以拉帕替尼为对照,BAF-0711-A对乳腺癌细胞MB-45-3d有较强的增殖抑制活性,其IC50值为0.217μmol/L。

【Abstract】 The purpose of the present study was to isolate,purify and identify components with antitumor activity produced by Streptomyces sp. BAF-0711. The secondary metabolite was isolated and purified by macroporous resin column chromatography,silica gel chromatography,etc.; the chemical structure was identified by physicochemical properties and spectroscopic methods; The antitumor activity was detected by MTT assay. As a result,compound BAF-0711-A was isolated from the fermentation broth of Streptomyces sp. BAF-0711 and was identified as bafilomycin A1 by spectroscopic methods. MTT assay showed that BAF-0711-A had strong growth inhibitory activity compared with lapatinib on MDAMB-453 cell,with IC50 value of 0. 217 μmol / m L.
