枣类黄酮化合物的快速提取方法
A Rapid and Efficient Extraction Method of Flavonoids from Ziziphus jujuba Fruits
【摘要】 针对枣类黄酮化合物提取中存在的耗时长、含糖量高等问题,以"鲁枣1号"为试材,采用紫外分光光度计和高效液相色谱进行枣类黄酮化合物的快速提取方法研究。本研究从提取质量、化合物数量、提取时间等多方面综合比较溶剂超声浸提法、融冻微波破壁法和简化QuEChERS法的提取效果,确定枣类黄酮化合物的快速提取方法。研究表明:简化QuEChERS法可在30 min内有效提取枣类黄酮。具体步骤为:样品加水涡旋5 min,然后加乙腈振荡15 min,最后加NaCl离心分层,取上层乙腈相。整个过程历时30 min,可有效去除部分糖类物质。该方法简单、快速、有效。
【Abstract】 The objective of this study was to develop a simplified and rapid procedure for flavonoids extraction from jujube fruits. Flavonoids were extracted from the ripe edible fruits of jujube( Ziziphus jujuba ’ Luzao 1’) using three methods,which were ultrasonic extraction,microwave- frozen- melted disintegrating method and simplified Qu ECh ERS method. The extraction effects were comprehensively evaluated from quantity,quality and time by Uv- vis spectrophotometry and high- performance liquid chromatography( HPLC).The results showed that the simplified Qu ECh ERS method was optimal. Detailed procedures were as follows: the grinded jujube fruits in liquid nitrogen were eluted by water for 5 minutes,and then acetonitrile was added to oscillate for 15 minutes. After oscillation,Na Cl was added to the extracts. By centrifugal separation,the extracts were turned into two layers,the upper acetonitrile layer contained all the flavonoids. The entire procedure can be carried out in approximately 30 minutes,and effectively eliminate some saccharide. The simplified Qu ECh ERS method is easy,rapid and efficient.
