血必净注射液对脑挫伤大鼠脑细胞凋亡及Bcl-2、Bax蛋白表达的影响
Effects of Xuebijing Injection(血液净注射液) on Apoptosis of Brain Cells and Expressions of Bcl-2 and Bax Protein in Rats with Brain Contusion
【摘要】 目的:探讨血必净注射液对脑挫伤大鼠脑细胞凋亡及Bcl-2、Bax蛋白表达的影响。方法:将36只Wistar大鼠随机分为正常对照组、脑挫伤组和血必净注射液组;制备大鼠脑挫伤模型,TUNEL法测定脑细胞凋亡率以及间接免疫荧光法测定大鼠脑细胞Bcl-2、Bax蛋白的表达。结果:与正常对照组比较,脑挫伤组、血必净注射液组大鼠脑细胞凋亡指数明显升高,差异显著,具有统计学意义(P<0.01);与脑挫伤组比较,血必净注射液组脑细胞凋亡指数明显降低,差异具有统计学意义(P<0.05)。与正常对照组比较,脑挫伤组、血必净注射液组大鼠脑细胞Bcl-2蛋白表达显著下降,Bax蛋白表达显著上升,Bcl-2/Bax比值显著减小,差异具有统计学意义(P<0.05);与脑挫伤组比较,血必净注射液组大鼠脑细胞Bcl-2蛋白表达显著上升,Bax蛋白表达显著下降,Bcl-2/Bax比值显著增大,差异具有统计学意义(P<0.05)。结论:血必净注射液可以影响Bcl-2、Bax蛋白的表达,有效抑制脑挫伤大鼠的脑细胞凋亡,对钝性脑挫伤具有一定的疗效。
【Abstract】 Objective: To explore the effects of Xuebijing Injection on brain cells apoptosis and gene expressions of Bcl- 2 and Bax protein in rats with brain contusion. Methods: 36 Wistar rats were randomly divided into normal control group,brain contusion and Xuebijing Injection group,brain contusion models were prepared,brain cells apoptosis rate was detected by TUNEL method,and gene expressions of Bcl- 2 and Bax protein were determined by indirect immunofluorescence method. Results: Compared with normal control group,brain cells apoptosis index of brain contusion group and Xuebijing Injection group increased significantly,difference was statistically significant( P < 0. 01). Compared with brain contusion group,apoptosis index of Xuebijing Injection group was obviously lowered,difference was statistically significant( P < 0. 05). Compared with normal control group,the Bcl- 2 protein expression in brain contusion group and Xuebijing Injection group decreased significantly,and the Bax protein expression increased significantly,the Bcl- 2 / Bax ratio was significantly reduced,statistically significant difference( P < 0. 05). Compared with brain contusion group,the Bcl- 2 protein expression was raised significantly in Xuebijing Injection group,and the Bax protein expression decreased significantly,the Bcl- 2 / Bax ratio was significantly increased,statistically significant difference( P < 0. 05). Conclusion:Xuebijing Injection can affect the Bcl- 2,Bax protein expressions,effectively inhibit apoptosis of brain cells in rats with cerebral contusion,has certain curative effect on blunt brain contusion.
