虎杖对放射性肺损伤大鼠血浆TGF-βIL-6、ACE含量的影响
Influence of Huzhang( Pdygonum Cuspldatum) on Plasma TGF- β,IL- 6,ACE Contents in Rats with Radioactive Lung Injury
【摘要】 目的:观察虎杖对放射性肺损伤大鼠血浆TGF-β、IL-6、ACE的影响,探讨虎杖防治放射性肺损伤的作用机制。方法:随机将180只SD大鼠分为正常对照组、模型组、虎杖预防组、虎杖即日组(0 d)、虎杖28 d组、DXM治疗组,共6组,每组30只。除正常对照组外,其余各组均给予直线加速器全胸单次照射18Gy造模。虎杖预防组于照射前1 W开始灌服虎杖流浸膏,虎杖0 d治疗组于造模后第1 d开始每日予虎杖流浸膏灌胃,虎杖28 d治疗组于造模后第28 d开始每日予虎杖流浸膏灌胃,地塞米松治疗组造模后第1 d开始每日予地塞米松灌胃,用药4 W,4 W后予1 m L/100 g生理盐水灌胃,正常对照组和模型组予等容积的生理盐水灌胃。每组分别于1、2、3、4、6、8 W随机处死4只大鼠,心脏取血,采用ELISA法测定血浆中TGF-β、IL-6、ACE的变化。结果:1)在照射后第4 W和第8 W时大鼠血浆TGF-β升高明显,与正常对照组比较,差异有统计学意义(P<0.01);第4 W、第8 W虎杖预防组、虎杖即日治疗组大鼠血浆TGF-β含量显著低于模型组(P<0.05),DXM治疗组在第8 W时显著低于模型组(P<0.01),虎杖28 d组与模型组比较无差异;虎杖预防组、虎杖即日治疗组TGF-β含量在第1、2 W时明显低于DXM治疗组,差异有统计学意义(P<0.05),但第3、4、6、8 W差异无统计学意义(P>0.05)。2)与正常组大鼠比较,模型组大鼠血浆中IL-6在第4、6、8 W显著升高(P<0.01);虎杖预防组和虎杖即日治疗组上升趋势较模型组、DXM治疗组慢,在第4、8 W时与模型组比较有统计学意义(P<0.01),各时间点与DXM治疗组比较有显著差异(P<0.01);而虎杖28 d治疗组与模型组比较无差异,但第8 W时IL-6含量显著低于DXM治疗组(P<0.01)。3)模型组大鼠血浆中ACE含量下降明显,与正常对照组比较有显著差异(P<0.01);虎杖预防组与虎杖即日治疗组ACE含量先下降,第4 W后渐升高,但低于DXM治疗组,与模型组、DXM治疗组比较第4、6、8 W均有显著差异,有统计学意义(P<0.01);虎杖28 d治疗组与模型组比较无差异(P>0.05)。结论:虎杖可以降低照射后大鼠TGF-β和IL-6水平,并且能上调ACE的表达,对放射性肺损伤具有防治作用。
【Abstract】 Objective: To observe the influence of Polygonum Cuspidatum on plasma TGF- β,IL- 6 and ACE contents in radiactive lung injury rats,and explore its possible mechanism of preventive effect. Methods:( 1) 180 male SD rats were randomly divided into normal control group,model group,prevention of Polygoncem Cuspidatum group,Polygoncem Cuspidatum same day group,Polygoncem Cuspidatum 28 days group,dexamethasone treatment group,each group had 30 rats. Except the normal control group,other groups were given single irradiation linac in 18 Gy to build models. Prevention group began to gastric gavage with Polygonum cuspidatum extract 1 weeks before irradiation,the same day group began to gastric gavage with Polygonum cuspidatum onthe first day after modeling,while 28 days group began to gastric gavage on the 28 th days,Dexamethasone treatment group after first day with dexamethasone orally for 4weeks,4 weeks later 1 m L /100 g normal saline,normal control group and model group were given saline gavage. 4 rats of each group were killed randomly respectively at 1,2,3,4,6,8 weeks. Rats’ heart blood was taken to determine the concentration of plasma TGF- β,IL- 6 and ACE by ELISA method. Results: 1) Compared with normal control group,TGF- β in rats’ plasma increased significantly in fourth weeks and eighth weeks after irradiation( P < 0. 01),compared with model group,the content of TGF- β in prevention of Polygonum cuspidatum group,the same day group,dexamethasone treatment group were lowered,at the fourth weeks,eighth weeks that in prevention group,same day group was significantly lower than that of model group( P <0. 05),DXM treatment group was significantly lower than that in model group at the eighth week( P < 0. 01).Compared with the model group,28 days group has no difference. Compared with DXM group,TGF- β in prevention group,the same day group was significantly lower on the first and second weeks,the difference was statistically significant( P < 0. 05),but no significant difference at the 3,4,6,8 weeks( P > 0. 05). 2) IL- 6 in rats’ plasma after irradiation was significantly increased,there was significant difference compared with the normal control group( P < 0. 01). Compared with model and DXM treatment group,IL- 6 content in prevention group,the same day group was increased slowly,it had significant difference at fourth and eighth weeks( P <0. 01),there was significant difference at each time point with DXM treatment group( P < 0. 01),28 day group had no difference compared with model group,but at eighth weeks,the IL- 6 content was significantly lower than that of DXM group( P < 0. 01). 3) The contents of plasma ACE of rats decreased after irradiation,at each time point was significantly different with the normal control group( P < 0. 01); ACE content in prevention group,the same day group decreased,then gradually increased after the fourth weeks,but lower than that of DXM group,compared with the model group and DXM treatment group,that in 4,6,8 weeks was significantly different with statistical significance( P < 0. 01); 28 day group had no difference compared with model group( P > 0. 05).Conclusion: Polygonum Cuspidatum can reduce the content of TGF- β and IL- 6,upregulate the expesion of ACE,so,that has certain prevention on radioactive lung injury.
