大鼠PSI-IH脊髓撞击损伤模型的制备

【摘要】 目的采用PSI-IH脊髓打击器制备不同程度的大鼠脊髓损伤模型。方法取成年雌性SD大鼠15只,随机分为3组,每组5只。采用PSI-IH脊髓打击器建立不同打击力度(80kdyn,120kdyn和160kdyn)的脊髓损伤模型。采用开放场地实验(BBB评分),于术前1d、术后1d、3d、1周及之后的每周分别进行后肢的运动功能评分。在损伤8周后取损伤段脊髓标本,切片、苏木精-伊红染色观察其损伤程度。结果术前所有的动物BBB评分均达21分。术后1d和3d,均为0分。随后,各组动物的运动功能逐渐恢复,至4周后达到平台。80kdyn组评分最高,120kdyn组评分居中,160kdyn组评分最低。苏木精-伊红染色表明80kdyn组脊髓损伤面积比例最小,120kdyn组居中,160kdyn组最大,与各组的BBB评分结果呈反向关系。结论采用PSI-IH脊髓打击器可以成功制备不同程度的大鼠脊髓损伤模型。

【Abstract】 Objective To establish the spinal cord injury(SCI)model of difference degrees by using the PSI-IH spinal cord impactor.Methods Adult female SD rats were randomly divided into three groups,5rats in each group.SCI model was established by using the PSI-IH impactor with different force(80,120 and 160kdyn).Behavioral assessment was performed using the Basso,Beattie,Bresnahan(BBB)locomotor rating scale,The BBB score was evaluated at 1and 3days,then 1,2,3,4,5,6,and 8weeks after injury.Eight weeks after SCI,animals were sacrificed and the histological damage was observed by HE staining.Results The BBB score of all animals was 21 points before SCI and decreased to 0point at 1day and 3days after SCI.Over the following days,the locomotor performance was gradually improved and reached a plateau at 4weeks.The BBB score in 80-kdyn group was the highest,followed by 120-kdyn group and then the 160-kdyn group.HE staining showed that the area of injured spinal cord was smallest in 80-kdyn group,and was gradually increased in 120-kdyn group and then 160-kdyn group,indicating an inverse relationship with BBB scores.Conclusion The rat spinal cord injury model of different degrees can be successfully established by using the PSI-IH impactor.
