微透析采样考察麻黄-桂枝药对配伍对大鼠额叶皮层部位神经递质Glu和GABA水平的影响
Study on Glu and GABA release in prefrontal cortex of rats treated with Mahuang-Guizhi herb pair
【摘要】 目的考察麻黄-桂枝药对配伍后对大鼠额叶皮层部位Glu和GABA等氨基酸类神经递质水平变化的影响。方法采用液质联用分析技术(UPLC-ESI-MS/MS),建立测定大鼠脑透析液样品中Glu和GABA等氨基酸类神经递质含量的方法。分别灌胃(ig)给予大鼠生理盐水、麻黄碱、麻黄、和麻黄-桂枝药对3∶2配伍供试药。采用微透析采样技术,收集额叶皮层部位的脑透析液,比较不同给药组间Glu和GABA浓度的变化差异。结果成功建立了UPLC-ESI-MS/MS测定大鼠脑透析液中Glu和GABA含量的方法。与生理盐水组相比,麻黄碱组、麻黄组和配伍组的Glu、GABA水平均有不同程度的升高。麻黄-桂枝药对配伍后,麻黄碱诱导的Glu水平升高得到显著抑制。结论麻黄-桂枝药对配伍后可以抑制麻黄碱诱导的中枢兴奋性变化。
【Abstract】 Objective To study the Glu and GABA release in rat prefrontal cortex when treated with Mahuang-Guizhi herb pair.Methods Develop and validate the ultra-high-performance liquid chromatography tandem mass spectrometry( UPLC-MS/MS) method to determine the release of Glu and GABA in rat prefrontal cortex. The release of amino acid neurotransmitter in the prefrontal cortex were determined in 3 hours after administered saline,ephedrine,ephedra and Mahuang-Guizhi herb pair solutions. Results The ultra-high-performance liquid chromatography tandem mass spectrometry( UPLC-MS/MS) method to determine the content of amino acid neurotransmitter in prefrontal cortex have been developed and validated. The release of Glu and GABA were increased compared to saline group. The content of Glu was significantly suppressed when treated with a Mahuang-Guizhi herb pair. Conclusion Guizhi inhibited ephedrine-induced excitotoxicity when treated with Mahuang-Guizhi herb pair.
