基于RAPD标记竹节参及其近缘植物的品种鉴别

【摘要】 目的建立竹节参及其近缘人参属植物种间快速鉴别体系。方法采用RAPD技术获得DNA指纹图谱,根据图谱中特异性条带,绘制人工绘制品种鉴定图(MCID)。结果从16条随机引物中筛选出能稳定扩增并且具有特异性条带的4条引物构建DNA指纹图谱,共扩增出46条带,其中多态性条带42条,多态性比率为91.30%。利用其中的多态性条带,绘制出图谱关系分析图,构成竹节参及其近缘植物的人工绘制品种鉴定图。结论该方法快速简单,结果直观可靠,为竹节参及近缘品种的初步鉴别提供了一种有效可行的方法。

【Abstract】 Objective This study was aimed at establishing a system for rapidly identifying Panax japonicus and its related plant species. Methods DNA fingerprinting was obtained by the RAPD technique. According to the presence or absence of specific bands in the DNA fingerprinting,manual cultivar identification diagram( MCID) was drawn. Results After screening 4 random primers which could stably amplify DNA fragments and obtain specific DNA bands were selected from 16 random primers. DNA fingerprinting was constructed using the 4 primers. A total of 46 bands were amplified by the 4 random primers,42 bands of which were the polymorphic bands. The percentage of polymorphic bands reached 91. 30%. According to some polymorphic bands of DNA fingerprinting,map relationship analysis diagram was drawn,which constituted the MCID of Panax japonicus and its related plant species. Conclusion The RAPD- MCID method was simple and rapid,and the results were visual and reliable. It provides an effective method for the preliminary identification of Panax japonicus and its related plant species.
