缺血再灌注对大鼠腮腺导管细胞TGF-β1、Bcl-2表达的影响

【摘要】 目的探讨缺血再灌注损伤对大鼠腮腺导管细胞TGF-β1、Bcl-2表达的影响。方法 wistar大鼠经颈总动脉结扎造成腮腺缺血再灌注模型,应用免疫细胞化学及一般光镜技术,分别观察缺血0.5 h和l h,再灌注6 h(早期)、12 h(中期)和24 h(后期)腮腺导管上皮细胞的TGF-β1和Bcl-2的动态表达情况及一般形态学变化。结果 1.TGF-β1:缺血再灌注组较对照组TGF-β1表达明显提高(P<0.01),且随缺血时间的延长,TGF-β1的表达呈逐渐增高趋势;组间比较,缺血1 h比0.5 h组表达提高(P<0.01);再灌注各组间两两比较差异均无统计学意义(P>0.05)。2.Bcl-2:再灌注6 h组的表达较对照组明显增高(P<0.05),而再灌注12、24 h较对照组差异无统计学意义(P>0.05);组间比较,缺血时间0.5 h与1 h组、再灌注各组间的两两比较差异均无统计学意义(P>0.05)。3.形态学:与对照组正常形态比较,实验组在缺血再灌注早期开始出现腺泡细胞萎缩、坏死等形态学改变,中期明显,后期修复。导管上皮细胞变化较轻。结论缺血再灌注可明显提高大鼠腮腺导管细胞TGF-β1的表达;缺血时间越长,TGF-β1表达上调越明显;缺血再灌注使Bcl-2的表达在早期明显升高;而中晚期Bcl-2的表达无显著变化;缺血再灌注对TGF-β1和Bcl-2表达的影响可能与形态学变化有关。

【Abstract】 Objective To explore the effect of experimental perfusion after ischemia on the expression of TGF- β1 and Bcl-2 in rat parotid gland ductal epithelial cells. Methods Ninety Wistar rats were subjected to ligation of bilateral neck arteries to establish the models of perfusion after experimental parotid gland ischemia. The dynamic expression and morphological changes of TGF- β1 and Bcl- 2 in parotid gland ductal epithelial cells at 0. 5 h and 1 h during ischemia and 6 h( early stage),12 h( middle stage) and 24 h( late stage) during perfusion were observed by using SP immunohistochemistry stain and light microscope( LM). Results The expression of TGF- β1 in the ischemia and perfusion groups was significantly increased as compared with the control group( P < 0. 01); moreover,the expression of TGF- β1 showed a gradually increasing tendency as the ischemia time prolonged. The expression of TGF- β1 in the 1h ischemia group was significantly higher than that in the 0. 5 h ischemia group,but it did not show statistically significant differences among perfusion groups of different time points( P > 0. 05). The expression of Bcl- 2 in the 6 h perfusion group was significantly increased as compared with the control group( P < 0. 05),but no statistically significant differences were found between the control group and the 12 h or 24 h perfusion groups( both P > 0. 05).Meanwhile,no statistically significant differences were found in the expression of Bcl- 2 between 0. 5 h ischemia group and 1 h ischemia group as well as among the perfusion groups( all P > 0. 05). At the early stage of parotid injured of ischemia and perfusion,the parotid acinar cells of the experimental groups showed morphological changes as compared with the normal acinar cell morphology of the control group,and the changes became more obvious at the middle stage. At the late stage the experimental groups underwent repair. The morphological changes of parotid gland ductal epithelial cells were milder. Conclusions The expression of TGF- β1 in rat parotid gland ductal epithelial cells is significantly increased under the condition of experimental perfusion after ischemia; moreover,the expression is significantly up- regulated as the ischemia time prolongs. The expression of Bcl-2 is significantly increased at early stage under the condition of experimental perfusion after ischemia,but no significant changes are found in the expression at middle and late stages. The impact of experimental perfusion after ischemia on the expression of TGF- β1 and Bcl- 2 may be correlated with the morphological changes of parotid gland ductal epithelial cells.
