HPLC-ELSD/HPLC-TOF/MS法分析大豆磷脂酰胆碱的分子种
Analysis of soybean phosphatidylcholine molecular species by HPLC-ELSD/HPLC-TOF/MS
【摘要】 目的运用HPLC-ELSD/HPLC-TOF/MS法对大豆磷脂酰胆碱(phosphatidylcholine,PC)的分子种进行定性及定量研究。方法色谱分离采用Phenomenex Luna C18(2)(250 mm×4.6 mm,5μm)色谱柱,流动相为甲醇-乙腈-水(体积比为90∶5∶5),柱温:40℃,流速:1.0 L·min-1,ELSD参数:载气流速为2.0 L·min-1,漂移管温度:72℃。质谱定性采用飞行时间质谱,ESI源,正离子模式,质量扫描范围为501 500。结果该方法能很好的分离大豆磷脂酰胆碱的分子种,并通过飞行时间质谱鉴定,自制磷脂酰胆碱所含的主要分子类型有18∶3/18∶3-PC、18∶2/18∶3-PC、18∶2/18∶2-PC、16∶0/18∶2-PC、18∶1/18∶2-PC和18∶0/18∶2-PC,经归一化法测得18∶2/18∶2-PC含量为73.9%。结论该方法具有高分辨率、高灵敏度的特点,为大豆磷脂酰胆碱质量控制的深入研究奠定了良好的基础。
【Abstract】 Objective To quantify and identify the soybean phosphatidylcholine molecular species by HPLCELSD / HPLC-TOF / MS. Methods The separation was performed on a Phenomenex Luna C18( 2) reverse phase column( 250 mm × 4. 6 mm,5 μm). The mobile phase consisted of methanol-acetonitrile-water( V∶V∶V =90∶5∶5). The flowrate was 1. 0 m L·min- 1and the temperature of column was 40 ℃. The nebulizing nitrogen gas flowrate and the temperature of the drift tube of the ELSD were 2. 0 L·min- 1and 72 ℃. Time-offlight mass spectrometer( TOF / MS) and electrospray ion source( ESI) were applied in qualitative analysis under positive ion mode,and mass scan range was m / z 100- 1 000. Results All molecular specieed could be separated by HPLC-ELSD and identified by HPLC / MS, and the content of each molecular specieed phosphatidylcholine could be also determined. The determining result on self-made PC by this method shows there are six molecular species phosphatidylcholine,they was 18∶ 3 /18∶ 3-PC,18∶ 2 /18∶ 3-PC,18∶ 2 /18∶ 2-PC,16∶ 0 /18∶ 2-PC,18 ∶ 1 /18 ∶ 2-PC and 18 ∶ 0 /18 ∶ 2-PC,and the content of 18 ∶ 2 /18 ∶ 2-PC was about 73. 9%.Conclusions This method has the characteristics of high resolution and sensitivity,which paves a way for the further studies of quality control of phosphatidylcholine.
