百合枯萎病病原菌的分离、鉴定及致病性研究

【摘要】 百合枯萎病是危害百合生长的重要病害。从3种不同杂种系的4个百合品种(亚洲百合Avelino,东方百合Limpopo和Topstan,OT系列百合Bonbini)的病变鳞茎中分离纯化真菌,经柯赫氏法则证病后,对分离菌进行形态学及分子生物学双重鉴定,在4种不同品种的百合病变鳞茎中鉴定出尖孢镰刀菌(Fusarium oxysporum)、三线镰刀菌(F.tricinctum)、串珠镰刀菌(F.moniliforme)和链格孢菌(Alternaria alternata)4种真菌均为病原菌。对亚洲百合Red Yunjing和东方百合Jumpy做致病性试验,结果表明:4种病原菌的致病程度强弱依次是尖孢镰刀菌、三线镰刀菌、链格孢菌、串珠镰刀菌。抗性分析结果表明:亚洲百合比东方百合对尖孢镰刀菌的抗性强。利用石蜡切片、苯胺兰染色以及荧光显微镜观察,了解菌丝在鳞茎和根部的侵染过程,为进一步从细胞水平研究百合枯萎病致病机理提供了理论依据。

【Abstract】 Lily blight is one of the important problems of hazard Lilies growth. Separation and purification of fungal from lesions lily bulbs of three kinds and four different varieties of hybrids. They are Asian lilies Avelino, oriental lilies Limpopo and Topstan, OT series lily Bonbini. After Koch’s postulates disease certification, we identified the four kinds of fungi through morphology and molecular biology. Results showed that they were Fusarium oxysporum, F.tricinctum, F. Moniliforme and Alternaria alternata, which were lily blight pathogen. Red Yunjing and Jumpy pathogenicity test results showed that pathogenic degree of strength followed by,Fusarium oxysporum, Fusarium tricinctum, Alternaria alternate, Fusarium moniliforme. The results showed that resistance was stronger than the East Asian lilies lily. Use paraffin, aniline blue staining and fluorescence microscopy to understand mycelium infection process in bulbs and roots, the anatomy of scales for disease pathogenes is further withered lily from the cellular level to provide a theoretical basis.
