固体基质中羊肚菌菌丝生物量的ELISA双抗体夹心检测法
Measurement of Morchella esculenta Mycelial Biomass in Soil Usinga Double Antibody-based ELISA Sandwich
【摘要】 为考察羊肚菌(Morchella esculenta)在固体培养基质中菌丝生物量,以液体培养的纯菌丝为标准,用抗羊肚菌菌丝抗原的鼠源单克隆抗体(C6A8)及兔源多抗(TM02)建立了羊肚菌菌丝生物量的酶联免疫吸附(ELISA)双抗体夹心检测法。该方法检测羊肚菌菌丝生物量的线性范围为2.58×102~6.6×104 mg/L,线性回归方程为Y=0.0949 X-0.0338,R2=0.9943;不同抗原浓度实验反应证明方法具有很好的重现性;这个方法用于黑脉羊肚菌(M.angusticeps)、尖顶羊肚菌(M.conica)、宽圆羊肚菌(M.rotunda)、金针菇(Flammulina velutipes)、黑木耳(Auricularia auricula)、金顶侧耳(Pleurotus citrinopileatus)的反应结果显示方法对羊肚菌属的供试菌株具有很好的特异性,其它种类的供试菌株不能引发特异的免疫反应。按照方法检测野生羊肚菌(M.esculenta)子实体周围土样,结果土壤基质中羊肚菌菌丝含量为0.75mg/g。
【Abstract】 A double antibody-based enzyme-linked immunosorbent assay(ELISA)sandwich to measure Morchella esculenta mycelial biomass in soil is described.A standard curve(correlation coefficient>0.99)was first constructed using a mycelium sample prepared from M.esculenta,strain 51589,mouse monoclonal antibody(C6A8)and rabbit polyclonal antibody(TM02).Detection levels ranged from 2.58×102 mg/L to6.6×104 mg/L and the method exhibited high reproducibility and specificity(strong positive reactions were obtained only with mycelium and fruit bodies of M.esculenta 51589 and mycelium of three other Morchella species).Using this method,soil taken from immediately below a fruit body was found to contain 0.75mg/g mycelium.
